Experiments were performed aiming at establishing optimal conditions for demonstrating the activity of dehydrogenases in Epon-embedded, semithin sections. Small fragments of the nervous tissue were incubated 1, 4, 10 and 24 h 37, 20 and 4 degrees C in the presence of NBT or TC NBT. The incubated fragments were embedded in Epon and semithin sections prepared thereof. The activities of succinate dehydrogenase and of alpha-glycerol dehydrogenase was evaluated. The experiments have shown that semithin, Epon-embedded sections may be used for demonstration and cytological localization of respiratory enzymes. Various experimental conditions were tried. Optimal results obtained when unfixed tissue fragments were incubated for 24 h at 4 degrees C.

15 ditetrazolium salts were examined to prove their qualities for histochemical techniques. The succinate dehydrogenase, the lactate dehydrogenase and the diaphorase I in hearts, muscles, livers, kidneys and brains of rats were demonstrated for it. The results show that NBT is the best allround tetrazolium salt for the histochemical demonstration of dehydrogenases. For the study of special questions it is suitable to use other tetrazolium salts, too.

The use of cinnamyl nitroblue tetrazolium chloride (DS-NBT) in dehydrogenase experiments (lactic dehydrogenase, succinic dehydrogenase, nicotinamide adenine dinucleotide diaphorase) and 3,3'-diaminobenzidine tetrahydrochloride (DAB) in cytochrome oxidase experiments indicated that mitochondrial oxidoreduction reactions from nicotinamide adenine dinucleotide to cytochrome oxidase are located on the inner mitochondrial membrane in the outer compartment and the intracristate spaces. These reactions behave according to the chemiosmotic hypothesis. The cochlear hair cell mitochondria are cytochemically indistinguishable from free liver mitochondria. The heterogeneous mitochondrial staining pattern is related to the osmolarity of the incubation media, solubility of the enzymes and pH of the medium, but not to the fixation method.

In a boy aged 8 years suffering from chronic granulomatosis of childhood necrotizing pneumonie and small pulmonary tuberculoid granulomas containing filaments of moulds were found at autopsy. Necrotizing leucocytic granulomas were present in the liver, spleen and the lymph nodes. All the organs showed aggregates of histiocytes containing yellowish cytoplasmic deposits of lipogment surrounded by a high acid phosphatase activity. The NBT-reduction leucocytic tests was repeatedly negative in vivo. The activities of NADH- and NADPH-tetrazolium reductase and succinate dehydrogenase in the tissues were histochemically normal.

In order to supply the tetrazolium salt procedure to flow cytometry we have synthesized monotetrazolium salts which are converted into fluorescent formazans on reduction. In this preliminary study we investigated the "oxygen sensitivity" of four of our compounds and compared them to the commercially available nitrotetrazole blue (NBT). The amount of formazan produced by Ehrlich ascites tumour cells via the succinic dehydrogenase reaction during oxic and anoxic incubations was determined by elution technique. Under oxic conditions no NBT formazan but much cyano-di-chlorophenyl formazan (==CCPC) was generated. The yields of this formazan differed only slightly between oxic and anoxic incubations. Twice as much of the amount of the other three formazans cyano-ditolyl- (==CTC, Stellmach 1984), cyanodiphenyl- (==CPC), and cyano-dianisyl tetrazolium chloride (==CAC), were produced in nitrogen atmosphere as in oxygen. The amount of all formazans investigated increased both in oxic and in anoxic conditions if cyanide was present. All cyano-aryl formazans have the maximum of absorbance between 430 and 470 nm and fluoresce in the spectral range above 580 nm with CTC formazan being the brightest. The tetrazolium salts, i.e. in unreduced state, did not fluoresce in the visible part of the spectrum. The formazans could be applied to flow cytometry. After staining the nuclear DNA with the fluorochrome DAPI, we obtained detailed two-parametric distributions correlating the amount of formazan per cell with the DNA content. The results are a step to establish a method for automated identification of malignant cells.

The new calcium antagonist anipamil (1,7-bis-(3-methoxyphenyl)-3-methylaza-7-cyano-nonadecane) exhibited a pronounced protective effect against isoprenaline-induced myocardial necrosis in rats. Anipamil was administered in single doses of 10 or 20 mg/kg daily for 4 days. 30 mg/kg isoprenaline was given by subcutaneous injection on the 3rd and 4th days of the study. The protective effect of anipamil was assessed by histological investigations, and its effect on the activity of the enzymes succinate dehydrogenase, NADH-NBT reductase, acid phosphatase and glucose-6-phosphate dehydrogenase in experimentally-induced myocardial damage was assessed quantitatively by microphotometry. The protective effect of anipamil against isoprenaline-induced myocardial necrosis was definitely dose-dependent: 10 mg/kg anipamil exhibited a partial protective effect, whilst 20 mg/kg anipamil protected the heart completely.

A histochemical technique was developed for the quantitative determination of succinic dehydrogenase (SDH) activity in muscle cross-sections using 1-methoxyphenazine methosulphate (mPMS) as the exogenous electron carrier, and azide as an inhibitor of cytochrome oxidase. The optimal composition of the incubation medium for the SDH reaction was determined. This histochemical procedure was compared to one using phenazine methosulphate (PMS) instead of mPMS and cyanide instead of azide. The substitution of mPMS and azide resulted in a substantial decrease in the non-specific reduction of nitroblue tetrazolium (NBT; the reaction indicator), i.e., 'nothing dehydrogenase' activity. With mPMS and azide in the reaction medium, the production of NBT formazan was linear for at least 9 min during the enzymic reaction. This compared to a non-linear reduction of NBT during the initial stages of the reactions (SDH and 'nothing dehydrogenase') when using PMS and cyanide. The use of both mPMS and azide also eliminated the production of NBT monoformazan which occurred with PMS and cyanide. This procedure was shown to meet various criteria established for the quantification of histochemical reactions.

Possible protective effects of Allium sativum and Crataegus--alone and in combination--on isoprenaline (isoproterenol)-induced heart, liver and pancreas damage were studied using rats as test animals. Pretreatment with Allium sativum alone, or in combination with Crataegus, resulted in protective effects on isoprenaline-induced damage of heart, liver, and pancreas. These effects proved to be dose-dependent. The following parameters were used to evaluate the protective effect: Clinical signs, qualitative histological and histoenzymatical findings, as well as quantitative microphotometric determination of enzymatic activities of succinate dehydrogenase, NADH-NBT reductase, acid phosphatase and glucose-6-phosphate dehydrogenase in cardiac, hepatic and pancreatic tissues. The underlying mechanisms are discussed. The results suggest that Allium sativum, resp. Allium sativum plus Crataegus exert a pronounced protective effect.

1-Methoxy-5-methylphenazinium methyl sulfate (MPMS) was tested for use as redox mediator in histochemistry of dehydrogenases. Aqueous solutions of MPMS are absolute lightstable. Like PMS or Meldola Blue the succinate dehydrogenase reaction was increased by MPMS. In an non-enzymatic NADH-NBT-test the reaction rate of MPMS exceeds the rates of PMS or Meldola Blue.

We studied energy metabolism in blood lymphocytes of Yakut ground squirrels Spermophilus undulatus in active state and during hibernation. Activities of succinate dehydrogenase (SDH) and lactate dehydrogenase (LDH), marker enzymes of mitochondrial respiration and glycolysis, were measured by a cytobiochemical method based on quantitative assessment of a product of NBT reduction to diformazan in blood lymphocytes immobilized on glass. To measure SDH and LDH activities, cytobiochemical staining of immobilized cells was performed with succinate, lactate, and NAD. In the state of hibernation, SDH activity decreased by 3 times and LDH activity decreased by 10 times or more. These results suggest that the decrease in metabolic activity in lymphocytes of ground squirrels during hypothermia is associated with inhibition of glycolysis, rather than with mitochondrial energy supply.
