The time-course of the ultrastructural changes and activities of 6 marker enzymes of subcellular particles (
succinate dehydrogenase
, beta-glucosidase, beta-N-acetylglucosaminidase, acid RNAse, glucose-6-phosphatase and
5'-nucleotidase
) has been studied in the liver, spleen and thymus in rats administered T-2 toxin (mycotoxin produced by some Fusarium species). A pronounced difference in the effect of T-2 toxin on the organs has been found. In the liver, the toxin induced a destruction of rough endoplasmic reticulum membranes, reduced ribosome number and progressively decreased activities of most enzymes. In the spleen, early and significant ultrastructural disturbances of all the cell membrane components and simultaneous lysosomal activation were observed. The changes in the thymus were characterized by a fast development of cell hydratation, organelle swelling and necrosis of some thymocytes with parallel increase in repair processes, infiltration by phagocytes and a selective activation of lysosomal hydrolases in the end of experimental time (72 h.). The results obtained emphasize an importance of cellular and subcellular membrane alterations in the mechanism of T-2 toxin action.

To define reproducible conditions for the homogenization of small-intestinal biopsy samples, tissue homogenization has been studied by the use of three different homogenizers. Tissue samples of increasing wet weights (0.5-10.8 mg) were homogenized in a fixed volume (1 ml) before DNA and protein were determined. The DNA to protein ratio was calculated for all wet weights and used as a measure for reproducible homogenization. The minimum tissue wet weight needed for analysis (2 mg) was determined from the values obtained for the DNA to protein ratio. Highly sensitive techniques are described in detail for the assay of brush border (maltase, lactase, sucrase, neutral alpha-glucosidase, alkaline phosphatase, gamma-glutamyl transferase, leucyl-beta-naphthylamidase), basolateral membrane (

The optimal dilution of tissue homogenate for the assay of the various enzymes has been determined to enable the determination of a maximum number of enzymes in each homogenate. Microsomal vesicles prepared from rat brain contain a Na+-Ca2+ exchange transport system capable of accumulating Ca2+ in a time- and temperature-dependent manner. The Ca2+ accumulated by these vesicles was released by the Ca2+ ionophore A23187 but not by EGTA. The Km value for Ca2+ uptake was 23 microM with a maximal velocity of 21 nmol Ca2+/mg per min. Ca2+ uptake was significantly inhibited by La3+, Sr2+, Mn2+ and Ba2+ and to a lesser extent by Mg2+. 45Ca2+ accumulated by Na+-dependent uptake could be released by 40Ca2+, indicating the presence of a Ca2+-Ca2+ exchange activity in the microsomes. Ca2+-Ca2+ exchange was stimulated in Li+- and K+-containing media as compared to choline+ media. Microsomes also catalyzed ATP-dependent Ca2+ uptake (in the absence of Na+ gradient). The Ca2+ sequestered by this mechanism could be released by extravesicular Na+, indicating that both the ATP-dependent and the Na+-dependent Ca2+ uptake systems are present in the same membrane. activity or oligomycin-azide-dinitrophenol sensitive ATP-dependent Ca2+ uptake. Thus, the Ca2+ accumulation observed was not due to contaminating mitochondria. The preparation was enriched for

Androgen binding activity was evaluated in different subcellular particulate fractions obtained by differential centrifugation of 32-day-old rat seminiferous tubules homogenates. After eliminating heavy particles by centrifugation at 4300 g during 4 min in 0.25 M sucrose buffer, a 27,000 g pellet was obtained and layered on 1.05 M sucrose buffer. The relatively light particulate interface (LPF) formed during centrifugation at 27,000 g 60 min, showed the highest androgen binding activity among particulate fractions. In LPF, concentrations of sites were estimated to be 328 +/- 54 fmol per mg proteins and affinity constant 0.78 +/- 0.23 10(9) M-1. Heat stability, steroid specificity, affinity constant and rate of dissociation were similar to the well known androgen binding protein, ABP. Presence of ABP or a similar protein in subcellular particles might play a role in the mechanism of action of androgens in seminiferous tubules of maturing rats.

Two enriched plasma membrane subfractions were obtained from syncytiotrophoblast isolated from human placenta. They were isolated from a "crude" plasma membrane fraction at the buffer-24% and 24-30% (w/w) sucrose interfaces of a sucrose gradient; another enriched plasma membrane fraction was isolated from the microsomal fraction at buffer-24% (w/w) sucrose interface and was similar to that isolated from the "crude" plasma membrane fraction at the same sucrose density. and alkaline phosphatase, the specific activity was twofold higher in the lighter than in the heavier subfractions. The activities of

Polyacrylamide-gel electrophoresis resolved the polypeptides of the plasma membrane subfractions into about 14 major protein bands; no differences were observed in the patterns of the two enriched plasma membrane subfractions derived from the "crude" plasma membrane fraction.

Epithelium in all regions showed acidpase staining. Oxidoreductases were found in all regions with variations within the epithelium. The epithelium of specific regions stained for alkpase and AMPase, while adjacent epithelium did not. We suggest that the alkpase and AMPase activities are associated with specific functions of the epithelium in these regions.

The effect of carrot extract on carbon tetrachloride (CCl4)-induced acute liver damage was evaluated. The increased serum enzyme levels (viz., glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, lactate dehydrogenase, alkaline phosphatase, sorbitol and glutamate dehydrogenase) by CCl4-induction were significantly lowered due to pretreatment with the extract.
