The presence of anions of phosphate (Pi), pyrophosphate (
PPi
), adenine nucleotides and sulfate greatly enhanced the production of superoxide radical (-O-2) by isolated guinea-pig macrophages. These anions, however, failed to enhance the production of -O-2 by the
xanthine oxidase
system, suggesting that they serve only as activators of -O-2 generating enzyme(s) located on the macrophage cell membrane. Many other common anions were ineffective in the macrophage system. In the presence of concentrations of Pi,
PPi
, adenine-5'-triphosphate (ATP) reported to be in the synovial fluid, -O-2 was produced efficiently and was inhibited by diclofenac sodium. These anions induced rat paw edema, maintained the swelling at least up to 6 h. The edema was suppressed partially by repeated injection of superoxide dismutase (SOD). High doses of sodium chloride and nitrate failed to maintain the swelling.

Coupled enzyme assays are described for measuring inorganic phosphates, organic phosphates and phosphate-liberating enzymes in biological material. The assays all determine Pi by its reaction with inosine, catalysed by nucleoside phosphorylase; this yields ribose 1-phosphate and hypoxanthine. The hypoxanthine is oxidized to uric acid by
xanthine oxidase
, and may be measured either by the absorbance of the uric acid, or by the formazan formed when a tetrazolium salt is used as the oxidant. The coupled enzyme assays are characterized by high sensitivity, quantitative utilization of phosphates and stoichiometric formation of the measurable products, measurement at pH 6.0-8.5, determination of phosphates within a single analytical step, and continuous measurement of phosphohydrolase activity in a corresponding rate assay. , does not seem to affect significantly the enzyme's catalytic activity. The steady-state parameters for the oxidation of several substrates by
xanthine oxidase
and oxygen have been determined. Formaldehyde differs from xanthine and other aldehydes in phi 2, the parameter describing the reaction with oxygen. Substrate inhibition has been studied at high concentrations of xanthine with oxygen as the electron acceptor. The inhibition is hyperbolic and uncompetitive with respect to oxygen. This is possibly due to rate-limiting product release from molybdenum(IV) being slower than from molybdenum(VI).

Oxygen-derived free radicals (ODFR) appear to be involved in the pathogenesis of arthritic disorders. In order to gain new insight on their role in the phenomenon and as a basis for a therapeutic approach, the effect of ODFR (produced by the
xanthine oxidase
-hypoxantine system) on hyaluronic acid, on two HA ester derivatives, and on pig articular chondrocytes was investigated. High M(r) HA (1.1 x 10(6)) and low M(r) HA (16 x 10(4)) were depolymerized by ODFR but the methyl and hydrocortisone esters of HA (HYAFF 2P50 and HYC13) turned out to be nearly unaffected. When articular chondrocytes were treated with ODFR, a rapid nucleoside triphosphate (NTP) depletion, a transient appearance of pyrophosphate (
PPi
), and an increase of phosphomonoester and diphosphodiester concentrations have been observed. The NTP depletion and the DPDE increase are related to the concentration of free radicals. Glyceraldehyde-3-phosphate accumulation during ODFR treatment suggests that ATP depletion can occur as a consequence of the blockage of glycolysis at the level of glyceraldehyde-3-P dehydrogenase. The hypothesis is presented that
PPi
can be produced from the pathway of the FAD-NAD (DPDE) biosynthesis and then either hydrolyzed by endogenous pyrophosphatases or precipitated in the form of insoluble calcium salts. Long-term treatment (16 h) with ODFR causes a loss of chondrocyte membrane integrity which can be revealed both by an increased free LDH activity and by the characteristic signal of free phospholipids in the 31P-NMR spectra. While high M(r) HA shows a significant protective activity for chondrocytes against ODFR action, low M(r) HA and ester derivatives do not. It is suggested that the therapeutic activity of HA ester derivatives can be ascribed to their in vivo hydrolysis products.
