The catalytic activity of phosvitin in Fe(II) oxidation and the addition of
iron
to transferrin were studied under various conditions. It was concluded that the Fe(II) oxidized by phosvitin would bind to apotransferrin, although an appreciable fraction of Fe(III) remained bound to phosvitin. Fe(III) also migrated from phosvitin to apotransferrin. This reaction was first-order with respect to Fe(III)-phosvitin concentration with a half-time (t1/2) of 10 min, and a first-order rate constant, k=0.069min-1, in 700 muM-phosphate buffer, pH 7.2, at 30 degrees C. The catalysis of the oxidation of Fe(III) by phosvitin was proportional to O2 concentration, and is quite different from the relative O2 independence of Fe(II) oxidation as catalysed by
ferroxidase
. A scheme for the mobilization and transfer of
iron
in the chicken, including the role of
ferroxidase
, phosyitin and transferrin, is presented.

In 9 groups of liver diseases, 11 serum protein parameters were studied. Prealbumin, retinol-binding protein, albumin, alpha-lipoprotein and Normotest (NT) were usually highly intercorrelated and separated best among groups with different types of functional impairment. Haptoglobin, C3 and total

-binding capacity were probably also sometimes reduced by restricted protein synthesis but appeared to be more easily affected by other factors, alpha1-antitrypsin was often increased in different liver diseases, but a pattern compatible with the "acute-phase reaction" was regularly present only in hepatic tumors. Bile retention was often accompanied by relatively high levels of
ceruloplasmin
, C3, and alpha-lipoprotein and by high NT values.

Nine samples of human
ceruloplasmin
[
iron

] prepared by different procedures have been examined for heterogeneity; gel electrophoresis showed that seven contained a number of components with molecular weights ranging from 20,000 to 130,000, and two contained largely a single component of molecular weight 130,000. Digestion of a single-component preparation with plasmin produced fragments with molecular weights similar to those found in the multicomponent preparations. Amino-terminal analysis, peptide mapping, and amino acid analysis showed that plasmin digestion generated a fragment of 20,000 molecular weight, which corresponded to a component present in a multicomponent
ceruloplasmin
preparation. The 20,000 molecular weight fragment appears to correspond to the so-called alpha-subunit or L-chain of human
ceruloplasmin
. Chemical evidence is thus provided that
ceruloplasmin
is a single-chain protein and that the so-called subunits are fragments. The 20,000 molecular weight fragment contains a single cysteine; amino acid sequence studies have shown that the sequence in the vicinity of this residue is similar to that around the single cysteine residue in plant plastocyanins and bacterial azurins, which are small, blue, copper-containing proteins.

The cellular immune response of a 17-year-old decerebrate male with acquired zinc deficiency was studied. He had been fed a commercial formula which contained 7.6 mg zinc per kilogram. His caloric intake had been inadequate as judged by his cachexia. A detailed pretreatment nutritional assessment (five separate observations) which included total serum protein and globulins, albumin, folate, vitamins A, B2, C,
ceruloplasmin
, and plasma zinc, copper,
iron
, and total
iron
binding capacity revealed that the patient was deficient only in zinc and calories. His plasma zinc was 41 +/- 5 microgram/d1 compared with our laboratory norm of 89 +/- 9 microgram/d1 for young adult males. Cellular immunity was assessed by delayed skin reactivity to dinitrochlorobenzene and by in vitro lymphocyte transformation studies. Before zinc therapy the patient rendered a negative skin reaction to dinitrochlorobenzene, and the ability of his lymphocytes to undergo blast transformation in response to mitogen stimulation was significantly depressed with a stimulation index of 4.7 +/- 0.8 as compared with 139.1 +/- 77.3 for controls. Within 3 weeks after zinc therapy (22.7 mg zinc per day) he demonstrated a positive delayed skin reaction to dinitrochlorobenzene and a normal lymphocyte response stimulation index = 205.5 +/- 42.6 versus 199.3 +/- 58.2 for control). In addition, a pretreatment facial seborrhea and a decubitus ulcer rapidly healed.

The blood, spleen and liver of mice were examined by means of electron spin resonance (e.s.r.), throughout the course of myeloid leukaemia induced by intravenous injection of leukaemic spleen cells. In blood, marked increases in the concentrations of
iron
transferrin and
ceruloplasmin
occurred within the first 3 days after injection. In the spleen, changes in the concentrations of paramagnetic copper and
iron
complexes were detectable by about the 5th day, before any measurable splenic enlargement, whilst in the liver changes were detectable by about the 8th day. The changes occurring in blood, spleen and liver during the development of leukaemia appear to be related and they are discussed in terms of
iron
transport.

) the blue Cu-protein of vertebrate plasma, possesses significant oxidase activity towards Fe(II) and numerous aromatic amines and phenols. Its
ferroxidase
activity has led to the discovery that it is a molecular link between copper and
iron
metabolism. Ceruloplasmin mobilizes
iron
into the plasma from
iron
storage cells in the liver. An additional role of Cp may be as a contributor to the regulation of the balance of biogenic amines through its oxidase action on the epinephrine and the hydroxyindole series. Ceruloplasmin also serves as a major copper transport vehicle, comparable to transferrin for
iron
. Evidence is presented that the copper atoms of Cp are a prerequisite for copper utilization in the biosynthesis of cytochrome oxidase. The ability of Cp to release copper at specific cellular sites is believed to be related to its broad substrate spectrum of biological reducing agents. Thus Cp is a serum protein with several important functions, all of which are directly related to its oxidase activity.

Electron spin resonance (ESR) spectroscopy was used to examine changes in the concentration of paramagnetic metal ions in Yoshida tumours carried by female Wistar rats. Blood, spleen and lymph nodes from these animals were also examined by ESR. A decrease in the concentration of a paramagnetic species associated with mitochondrial activity, and marked increases in those thought to be associated with inflammatory or immune reactions and cell lysis, were observed in the tumours within one day of implantation. During development of the tumour, and during its regression after treatment with methylene dimethane sulphonate (MDMS), further changes were observed in the concentration of the species. These were dependent on the region of the tumour examined. In blood, development of the tumour produced an increase in
ceruloplasmin
and a decrease in
iron
-transferrin. An increase in spleen weight, as the tumour developed, was accompanied by a small decrease in the concentration of species with g-values of 6-0 and 4-3, which was reversed on regression of the treated tumour. The presence of metastases in the regional lymph nodes produced distinguishable changes in the ESR spectra.

Electron spin resonance spectroscopy has proved a useful and simple technique for the measurement of levels of
caeruloplasmin
and
iron
transferrin in whole blood from 50 patients with Hodgkin's disease. Those patients with clinically active disease show higher
caeruloplasmin
levels and lower
iron
transferrin levels than those with inactive disease. The results indicate that these tests are good indicators of the state of the disease and that serial measurement of these parameters may help in early prediction of clinical reactivity and in monitoring response to treatment. The combined information from
iron
transferrin and
caeruloplasmin
levels appears to be more predictive than that from the erythrocyte sedimentation rate and neutrophil alkaline phosphatase score.

Caeruloplasmin and

transferrin level were measured in blood of patients with non-Hodgkin's lymphoma in different stages of disease activity and compared with erythrocyte sedimentation rate and NAP level in the same samples. It was found that both
caeruloplasmin
level and sedimentation rate showed a slight increase in mean level in patients with active disease as compared with those in remission, particularly in the group of patients with poorly or undifferentiated diffuse disease. No difference was observed in levels of
iron
transferrin or NAP. Both
caeruloplasmin
and sedimentation rate showed occasional abnormal values in patients in remission but in most cases where both were elevated the patients subsequently entered a more active phase of the disease.

Electron spin resonance studies have been made of
caeruloplasmin
and
iron
transferrin levels in whole blood of healthy controls and patients with a variety of malignant conditions receiving various forms of treatment. A small difference was found in
caeruloplasmin
level between normal males and females, although normal females receiving contraceptive steroids had an elevated level. No difference was found in
iron
transferrin level. Various conditions increased the
caeruloplasmin
and some also decreased the
iron
transferrin level in patients with malignant disease. These included surgery and the approach of a terminal phase of disease. Once allowance for these factors was made, the remaining small differences in Cu and Fe between patients with either squamous cell carcinoma or breast cancer and controls appeared to have no clinical significance.
