We developed a direct, simple, and sensitive procedure for the simultaneous colorimetric assay of iron and copper in serum, using sodium dodecyl sulfate-ascorbic acid to dissociate iron and copper from transferrin and
ceruloplasmin
, respectively. We also use a new water-soluble reagent, 2-(5-nitro-2-pyridylazo)-5-(N-propyl-N-sulfopropylamino)phenol disodium
salt
(nitro-PAPS) and thioglycolic acid to eliminate interference from copper in the measurement of iron. Within- and between-run precisions of the present method were 2.5-2.8% for iron and 1.8-4.6% for copper. The proposed method is susceptible to interference by hemoglobin and lipemia, especially for the iron assay. Linear-regression analyses of results of the proposed method with those of the bathophenanthroline method for iron and of the atomic absorption spectroscopic method for copper correlated well (r = 0.996, Sy/x = 0.73 and r = 0.959, Sy/x = 1.11, respectively).

-sensitive rats were fed, in a 2 x 3 factorial design, one of six diets that contained one of three levels of copper (2.0 micrograms/g marginal, 12 micrograms/g adequate, or 50 micrograms/g supplemental) and either control (0.4%) or high (4%) levels of sodium. Diets were fed to the rats for 11 weeks. Rats fed the high sodium diets were characterized by high plasma copper concentrations and

activities compared with their respective control sodium rats. The magnitude of the sodium-induced rise in plasma copper and

Basing on the literature data and their own findings, the authors present the general biological concept of osteogenesis. The significance of copper compounds (
ceruloplasmin
) in bone tissue synthesis was revealed; the role of bound iron (transferrin) in the development and maintenance of the inflammatory process induced by opportunistic bacteria and in bone regeneration was confirmed. Analysis of clinical and microbiological findings in the patients with chronic posttraumatic osteomyelitis, coral uroliths, gastrointestinal diseases complicated by cholelithiasis has lead the authors to a suggestion that fecal microcenosis may become one of the criteria for estimating mineral metabolism in the body; the markers of this metabolic disorders are shifted levels of full-value E. coli, Lactobacilli, and, to a lesser degree, of L. bifidum. The presence of E. coli hemolytic variant also essentially contributes to these disorders, when its level surpasses by 2-3 orders the level of this microorganism in normal subjects and in patients whose mineral metabolism is virtually unchanged. When planning multiple-modality treatment of patients with locomotor system defects and disordered

metabolism, a physician should investigate intestinal biocenosis and prescribe agents to correct the detected disorders.

Cancer of the gastrointestinal tract (GIT) is very commonly encountered in Kashmir Valley of India. Serum copper and
ceruloplasmin
levels were investigated in 75 patients with GIT cancer, 42 non-GIT cancer and 75 normal healthy controls. The mean serum copper in the control group was 1,078 (S.D. 0,296) mg/L, while in patients with GIT and non-GIT cancers, it was 1,738 (S.D. 0,675) and 1,449 (S.D. 0,501) mg/L. Copper in both types of cancers showed significant increase (p less than 0.001). It was significantly higher in GIT cancers (p less than 0.01) as compared to non-GIT cancers. Ceruloplasmin in controls was 379 (S.D. 51) mg/L, while in GIT and non-GIT cancer patients the values were 363 (S.D. 75) and 364 (S.D. 77) mg/L, respectively. No significant difference could be found in either of the groups. In Kashmir Valley, the copper content in body tissues is likely to be on the high side because copper utensils are widely used by the general population. isolated from human donor blood was investigated. It was assumed that the imidazole group of histidine is functionally important for the above reactions. For Fe(II) the effect of the ionizeable group was observed during substrate binding to the
ceruloplasmin
molecule, whereas in the course of the adrenaline oxidation reaction it manifests itself during catalytic interaction of the substrate with the enzyme. The organic substrate can bind both to the protonated and to the non-protonated form of the enzyme. Fe(II) interacts only with the protonated form of the protein. In both cases, the rate-limiting step of the oxidase reaction is preceded by a single step, i.e., proton binding. The schemes describing the order of proton attachment in the course of the above reactions are proposed.

A cross-sectional epidemiological study was carried out among 141 male subjects exposed to inorganic manganese (Mn) in a Mn oxide and
salt
producing plant (mean age 34.3 years; duration of exposure, mean 7.1 years, range 1-19 years). The results were compared with those of a matched control group of 104 subjects. The intensity of Mn exposure was moderate as reflected by the airborne Mn levels and the concentrations of Mn in blood (Mn-B) and in urine (Mn-U). A significantly higher prevalence of cough in cold season, dyspnea during exercise, and recent episodes of acute bronchitis was found in the Mn group. Lung ventilatory parameters (forced vital capacity, FVC; forced expiratory volume in one second, FEV1; peak expiratory flow rate, PEFR) were only mildly altered in the Mn group (smokers) and the intensity and the prevalence of these changes were not related to Mn-B, Mn-U, or duration of exposure. There was no synergistic effect between Mn exposure and smoking on the spirometric parameters. Except for a few nonspecific symptoms (fatigue, tinnitus, trembling of fingers, increased irritability), the prevalence of the other subjective complaints did not differ significantly between the control and Mn groups. Psychomotor tests were more sensitive than the standardized neurological examination for the early detection of adverse effects of Mn on the central nervous system (CNS). Significant alterations were found in simple reaction time (visual), audioverbal short-term memory capacity, and hand tremor (eye-hand coordination, hand steadiness). A slight increase in the number of circulating neutrophils and in the values of several serum parameters (ie, calcium,
ceruloplasmin
, copper, and ferritin) was also found in the Mn group. There were no clear-cut dose-response relationships between Mn-U or duration of Mn exposure and the prevalence of abnormal CNS or biological findings. The prevalences of disturbances in hand tremor and that of increased levels of serum calcium were related to Mn-B. The response to the eye-hand coordination test suggests the existence of a Mn-B threshold at about 1 microgram Mn/100 ml of whole blood. This study demonstrates that a time-weighted average exposure to airborne Mn dust (total dust) of about 1 mg/m3 for less than 20 years may present preclinical signs of intoxication.

Patients with rheumatoid arthritis have altered protein patterns in their serum and synovial fluid which influences the antioxidant activity of these fluids. Rheumatoid serum has a higher antioxidant activity than control serum when ferrous and ferric ions stimulate membrane damage. The raised levels of

and the lower iron saturation of transferrin contribute to these differences. When membrane damage is stimulated by a copper

, rheumatoid serum does not show an increased antioxidant protection and has probably a lower protective activity than control serum. Attempts to damage

and transferrin with oxygen radicals were unsuccessful. However, prolonged incubations with trypsin reduced the iron-binding capacity of transferrin and decreased the

Severe copper deficiency was induced in rats by rearing nursing dams and their offsprings on a semisynthetic diet comprising all the requisite nutrients and trace metals except copper. The copper-deprived rats exhibited growth retardation, severe anaemia, loss of
caeruloplasmin
, decrease of cytochrome oxidase, accumulation of
salt
-soluble collagen and a drastic decrease in iron in plasma and liver. Apart from these characteristic signs of deficiency, a marked inhibition of protein synthesis was found to occur both in vivo and in cell-free liver preparations. The curtailed ability to carry out endogenously coded amino acid incorporation into protein contrasted with the unimpaired poly(U)-acid-directed phenylalanine polymerization. This inhibition pattern, as well as the attendant disaggregation of the liver polyribosomes, suggested that the primary biosynthetic lesion was located at the stage of peptide-chain initiation. Concurrently with this alteration there was a pronounced depletion of the hepatic ATP content, associated with a parallel depression of mitochondrial respiration and an enhancement of ATPase activity. Supplementation of the copper-deficient diet with a 2-4-fold excess of iron (relative to the standard diet) prevented growth retardation and anaemia and restored normal energy metabolism, as well as unimpaired protein-synthesizing capacity. The conclusion that these disturbances were primarily determined by the secondary iron deficiency was also borne out by the finding that similar alterations occurred in rats maintained on a copper-sufficient but iron-deficient diet. On the other hand, the iron-fortified diet failed to reverse the other signs of copper deficiency, namely the loss of
caeruloplasmin
, the diminished rate of cytochrome oxidase and the increase of soluble collagen. The interrelations between the various biochemical lesions induced by deprivation of copper or iron are discussed and the possible role of ATP depletion in determining the derangement of protein synthesis is considered.

The chromatographic behaviour of 27 different plasma proteins on fractionation of human plasma on immobilized Cibacron Blue F3-GA was studied. The column was eluted by using a three-step procedure. First, a low-molarity buffer (30 mM-H3PO4/Na3PO4, pH 7.0, I0.053) was used, then a linear
salt
gradient (0-1 M-NaCl in the buffer above) was applied, followed by a wash with two bed volumes of 1.0 M-NaCl. Finally, bound proteins were 'stripped' with 0.5 M-NaSCN. Up to 1 ml of whole plasma could be loaded per 5 ml bed volume. No denaturation of proteinase inhibitors or complement fractions was observed. The recovery of individual proteins ranged between 52 and greater than 95%. Enrichment of four individual plasma components (alpha 1-antitrypsin,
caeruloplasmin
, antithrombin III and haemopexin) was between 10-fold and 75-fold. These results indicate that chromatography on immobilized Cibacron Blue F3-GA can be a useful initial step in the purification of plasma proteins.
