Parallel determinations were made, of 10 individual serum proteins and of proteins in the bronchial secretion obtained by aspiration from 8 subjects that did not show manifest bronchopulmonary disease. By application of the formula suggested by Deuschl and Johansson it was found that 37 percent of IgG, 84,5 percent of IgA, 48,9 percent of transferrins, 15,2 percent of alpha-1-anti-trypsin and 11,3 percent of ceruloplasmin present in the bronchial secretion are synthetised in the bronchial mucosa itself, while the rest occur as a result of diffusion from the blood. Acid alpha-1-glycoprotein and haptoglobin from the bronchial secretion originate in the blood. IgM was evidenced in the bronchial aspirate in only 3 subjects, and alpha-2 macroglobulin was not found in any of the subjects.

Amino acid sequences of 8 plastocyanins, 8 azurins, stellacyanin, two regions in human ceruloplasmin (ferroxidase)--all of which proteins are known to bind a blue (type 1) copper--and subunit II of bovine mitochondrial cytochrome c oxidase were compared by statistical methods to assess similarities and derive possible evolutionary relationships. It is suggested that all of the examined proteins are monophyletic. The two ceruloplasmin partial sequences clearly demonstrate that this protein has undergone a duplication. A calculated most parcimonious phylogenetic tree shows the divergence of the azurin and plastocyanin ancestor to be the earliest event. Stellacyanin and later the blue oxidase (ceruloplasmin) evolved from the plastocyanin branch, which the cytochrome c oxidase subunit evolved from the azurin ancestor.

2 studies, A and B, were conducted to determine the metabolic effects of oral contraceptives (OCs). The subjects were healthy nonsmokers (17 to 27 years old, 1.52 m. to 1.75 m. in height, and 50 kg. to 60 kg. in weight) who were not undergoing any therapy. Controls did not have any previous history of hormonal therapy. In Study A, biochemical data from 17 women who were not on OCs were compared with those of women taking pills containing either 30 ug of ethinyl estradiol (EE) and 150 ug of D. norgestrel (18 women) or 50 ug. of EE and 250 ug. of D. norgestrel (9 women). In Study B, 8 women were studied before and during 3 to 4 cycles of low-dosed OC therapy (30 ug. of EE and 150 ug. of D. norgestrel). In both studies, the 2 oral contraceptive dosage forms had similar metabolic quantitative and qualitative changes: both resulted in an increase in serum concentration of triglycerides (30 to 33%), B-lipoproteins (27 to 29%), and ceruloplasmin (75 to 90%), and a decrease in serum levels of antithrombin 3 (22 to 29%) and ascorbic acid (30 to 42%). Serum cholesterol and phospholipid concentration did not change. The proportion of serum cholesterol carried by a-lipoproteins (high density lipoproteins) did decrease (the change is much smaller than that seen in coronary heart disease) while that carried by B-lipoproteins (or low density and very low density lipoproteins) increased. A significant negative correlation was observed between serum concentrations of ascorbic acid and cholesterol; this is interesting as clinical and experimental evidence suggests that latent ascorbic acid deficiency leads to hypercholesteroleamia, while ascorbic acid supplements reduce plasma cholesterol levels. As 500 mg. daily of ascorbic acid supplements supposedly help maintain normal ascorbic acid levels in blood during OC use, they should perhaps be prescribed to pill users in this study.

Frequent blood transfusions are necessary in aplastic anemia because of decreased hematopoietic function. In order to maintain the hemoglobin level and to decrease the need for blood transfusion, human ceruloplasmin was used for the treatment of 73 patients with aplastic anemia. A marked beneficial effect was obtained in 16 cases. The treatment was moderately effective in 17 cases, slightly effective in eight cases, and ineffective in 32 cases. The usual dose of ceruloplasmin was 15 mg/day, but it was varied according to symptoms. Side effects were minimal when ceruloplasmin was administered by slow intravenous injection.

In summary, purified human ceruloplasmin inhibits several reactions mediated by superoxide anion in a fashion consistent with an ability to scavenge this free radical. It must be pointed out, however, that on a weight basis, the superoxide-scavenging activity of ceruloplasmin is substantially less than that of purified human erythrocyte superoxide dismutase. Nevertheless, since superoxide dismutase is almost exclusively an intracellular enzyme, ceruloplasmin probably represents the major circulating scavenger of superoxide anion radicals. The level of superoxide dismutase in human plasma has been reported to be 0.7 microgram/ml. It is not clear, however, how this was measured. We have found that concentrations of plasma exceeding 10% (v/v) interfere significantly with the assays routinely employed for detecting superoxide-scavenging activity. Consequently, we have not yet been able to quantify the superoxide-scavenging activity of either ceruloplasmin or superoxide dismutase in whole human plasma. Thus, we can only speculate that under conditions where levels of ceruloplasmin are markedly elevated, as during pregnancy, during acute infections, or in association with inflammatory diseases (such as rheumatoid arthritis), this acute-phase reactant may play a major role as a circulating scavenger of oxygen-derived free radicals.

Ceruloplasmin, the blue copper-protein of the blood plasma, is believed by some workers to be involved in the metabolic management of 5-hydroxytryptamine (serotonin) during pregnancy. 5-Hydroxytryptamine is abortifacient in experimental animals. By some authors, a role has been suggested for it in human abortion. The plasma concentrations of 5-hydroxytryptamine, copper and ceruloplasmin were measured in non-pregnant women, in normal early pregnancy and in cases of spontaneous abortion. Compared with normal early pregnancy, cases of inevitable abortion show lower values for both plasma copper and ceruloplasmin and higher values for plasma 5-hydroxytryptamine. The possible implications of these findings are discussed in view of the alterations in ceruloplasmin values in pregnancy and in the light of what is known of the pharmacology and metabolism of 5-hydroxytryptamine. It it believed that these alterations in plasma copper, ceruloplasmin and 5-hydroxytryptamine in cases of inevitable abortion are the effect, rather than the cause, of abortion.

The complete amino acid sequence has been determined for a fragment of human ceruloplasmin [ferroxidase; iron(II):oxygen oxidoreductase, EC 1.16.3.1]. The fragment (designated Cp F5) contains 159 amino acid residues and has a molecular weight of 18,650; it lacks carbohydrate, is rich in histidine, and contains one free cysteine that may be part of a copper-binding site. This fragment is present in most commercial preparations of ceruloplasmin, probably owing to proteolytic degradation, but can also be obtained by limited cleavage of single-chain ceruloplasmin with plasmin. Cp F5 probably is an intact domain attached to the COOH-terminal end of single-chain ceruloplasmin via a labile interdomain peptide bond. A model of the secondary structure predicted by empirical methods suggests that almost one-third of the amino acid residues are distributed in alpha helices, about a third in beta-sheet structure, and the remainder in beta turns and unidentified structures. Computer analysis of the amino acid sequence has not demonstrated a statistically significant relationship between this ceruloplasmin fragment and any other protein, but there is some evidence for an internal duplication.

LP, a saline extract of exudate-derived rabbit granulocytes, shown to be free of endotoxin contamination by sensitive LAL assay, can elicit brisk acute-phase responses in the rabbit. Following a single large intravenous dose of LP (875 mce) there is a brisk fall in serum iron at 8 hr and marked elevations in concentrations of CxRP, haptoglobin, fibrinogen, and ceruloplasmin and a lesser rise in sialic acid in the blood at 24 hr, which return toward baseline to varying degrees by 48 hr. When the crude LP solution is fractionated by column chromatography on Sephadex G75, all detectable acute-phase mediating activity elutes in a fraction (pool C) which contains all the pyrogenic activity and about 15% of the total eluted protein. Within the limitations of the methods employed, the acute phase stimulating activity and the pyrogenic activity of LP preparations appear to be closely linked.

Following extensive bowel resection, a young woman experienced severe malnutrition; subsequent administration of parenteral nutrition precipitated the copper deficiency syndrome. This consisted of hypocupremia, subnormal ceruloplasmin levels, anemia, and severe neutropenia. The bone marrow was megaloblastic, vacuolated, and sideroblastic; granulocytic maturation was not observed beyond the myelocyte stage. Copper sulfate therapy was followed by a marked reticulocytosis, increase in hematocrit, and recovery of neutrophils. Additional studies indicated that both serum and urinary erythropoietin values were low; serum activity increased after copper supplementation. Abnormal granulopoiesis was demonstrated using the in vitro granulocyte colony assay. The patient's granulcoytic stem cells were normal on two occasions; however, mixing studies showed that culture of the patient's copper-deficient marrow with her copper-deficient serum yielded significantly reduced numbers of granulocyte colonies. Thus, copper appears to be a necessary element for normal hematopoiesis; lack of this trace element may result in ineffective erythropoiesis and granulopoiesis.
