Although interaction of vitamin C, copper and iron have been studied in several species, little is known about these interactions in species which require the vitamin in the diet. Young male Hartley guinea pigs were fed a basal diet, or a basal diet and supplemented daily with vitamin C, p.o. Pharmacologic doses (25 mg per 100 g BW per day) of vitamin C resulted in two-to-three-fold decreases in liver copper, when compared with those receiving normal (0.5 mg per 100 g BW per day) intakes. Under conditions of vitamin C deficiency, serum copper and ceruloplasmin were elevated along with liver copper. Serum and hepatic iron levels, hepatic microsomal cytochrome P-450 and cytochrome b5, and blood heme parameters all appeared to be directly related to vitamin C intake, i.e. the iron and heme parameters increased as the vitamin dose increased. These data are consistent with the hypothesis that interaction between vitamin C, copper and iron influence normal heme formation through the oxidation/reduction of iron and/or by regulating iron absorption and availability at the gut level.

Molecular mechanisms of the trauma stress of different origins in children to some extent is related to the quantitative changes of blood superoxide dismutase, catalase, ceruloplasmin, transferrin, cytochromes b5, b558III, b558IV and suprol. Non adequate changes of antioxidant (superoxide dismutase, catalase, ceruloplasmin, transferrin) and newly discovered prooxidant (cytochromes b5, b558III, b558IV, suprol) metalloproteins, as well as serum and erythrocyte cytochrome b5 depend on characteristic and duration of trauma stress caused by extremity fractures in children. This is due to the different mechanisms of adaptation to trauma stress.

Changes in the contents of blood metalloproteins with prooxidant (plasma cytochromes b558 I and b558 II, erythrocyte membrane cytochromes b558 III and b558 IV, superoxide-producing plasma lipoprotein suprol, and cytochrome b5 from soluble erythrocyte fractions) and antioxidant activities (Cu,Zn-superoxide dismutase, catalase, ceruloplasmin, and transferrin) depended on the duration of hypokinesia (5, 10, and 15 days). The content of metalloproteins, particularly cytochrome b5 and ceruloplasmin, increased at the initial stage, but decreased at later stages of hypokinesia (except for cytochrome b5 concentration, which continued to increase).

Various stages of hypokinesia are marked by specific changes in the level and activity of endogenous metalloproteins with anti- (MAA) and prooxidant action (MPA). In particular, the level of MPA [cytochromes b5 and b558 (III + IV) and suprol] decreases in all stages (except for the level of cytochrome b5 exceeding that in the free control on the 15th and 30th day of hypokinesia). The superoxide production activity of suprol always exceeds that in the control, while the activity of cytochrome b558 (III) exceeds that in the control only on the 30th day of experiment. The level and activity of MAA [ceruloplasmin, transferrin (TF), superoxide dismutase (SOD), and catalase] decrease with the duration of hypokinesia (except for 15th day, when the level of TF and the activity of SOD exceed the values in the free control group). The administration of GABA and pyrrolidone decreases the extent of oxidative stress, which is manifested by leveling of the MAA and MPA content and activity.
