Ceruloplasmin is a copper containing serum glycoprotein. It is an acute-phase protein and its levels are increased in inflammation and in a number of experimental and human tumors. It is normally synthesized in the liver and not in fibroblasts. In this paper we present evidence that ceruloplasmin mRNA is synthesized in Balb/C 3T3 cells and its levels are increased about 3-fold in Rous-
Sarcoma
virus (RSV) transformed cells. The
ceruloplasmin
mRNAs from rat liver and RSV 3T3 cells have the same electrophoretic mobility.

Serum copper content and
caeruloplasmin
activity were measured in 595 Strain A male mice. The mice carried either benzo(a)pyrene induced fibrosarcoma, transplanted
sarcoma
-180, Dalton's lymphoma, or Ehrlich carcinoma. The serum copper concentration was raised in all the tumours when compared with normal controls. The observed fall in activity of
caeruloplasmin
during malignancy may be related to defective iron mobilization.

Transplantation of Yoshida
sarcoma
(solid type) and Zajdela ascites hepatoma tumors in rats induces a biphasic change in the concentration of the following five acute-phase proteins: alpha-1-acid glycoprotein; alpha-1-antitrypsin; haptoglobin; hemopexin; and
ceruloplasmin
. These proteins and other plasma proteins were quantitated by two-dimensional immunoelectrophoresis relative to normal serum concentrations. The elevation of most of these acute-phase proteins was greater in the second phase, during which serum levels increased continuously as the tumor burden increased until the animals died. The increase in haptoglobin concentration during the second phase was much higher in rats bearing Yoshida
sarcoma
than in rats bearing Zajdela tumors. Rats receiving irradiated tumor cells showed neither tumor growth nor second-phase protein changes. Significant increases in uptake of 3H-amino acids by isolated perfused livers of tumor-bearing rats provided evidence for an increase in the hepatic synthesis rates of the acute-phase proteins. Removal of the solid tumor resulted in a gradual decrease of acute-phase protein concentrations with concomitant increase in serum albumin concentration. These alterations in serum acute-phase proteins during tumor growth and after removal of the tumor may make their use attractive as biological markers of the response of the tumor-bearing animal to its tumor.

Human
ceruloplasmin
was shown to have a restorative effect on the decrease of delayed hypersensitivity in
sarcoma
-180-bearing mice and of helper T-cell activity in cell-free. Ehrlich cancerous ascitic fluid-treated mice. Furthermore, it was also shown that
ceruloplasmin
augments the in vivo generation of alloreactive cytotoxic cell which consist of cytotoxic T-lymphocytes, cytotoxic macrophages and K cells.

High levels of serum
ceruloplasmin
oxidase activity were detected in U strain isogenic male rats bearing a syngeneic
sarcoma
in all phases of tumoral evolution. Normal levels of
ceruloplasmin
were only observed after a single i.p. dose of cyclophosphamide (100 mg/kg). However subsequent weekly administration of the drug did not prevent the return of hyperceruloplasminemia in the tumor hosts. Normal rats treated similarly with cyclophosphamide presented a fall in the enzyme activity after the 1st dose.

Human
ceruloplasmin
was found to have a neutralizing effect against the toxohormone activities of the basic protein isolated from Ehrlich carcinoma cells; this basic protein decreases the levels of serum iron and liver catalase activity upon intraperitoneal injection into mice and shows direct cytotoxicity toward normal mouse lymphocytes and macrophages. Furthermore, it was also shown that
ceruloplasmin
has antitumor activity against
sarcoma
-180 cells implanted in ICR mice.

To specify mechanisms of paraneoplastic alterations of redox-status of tissues in experimental malignant tumor growth, we investigated electronic paramagnetic centers of blood, skeletal muscle and liver with electronic paramagnetic resonance (EPR) method. We also studied the concentration and activity of antioxidant enzymes. Our experiments on adult white male rats of mixed population with
sarcoma
C-45 and mice with Ehrlich carcinoma have shown that malignant tumor growth leads to enhanced lipid peroxidation (LPO): production of potent LPO promoters - Fe2+, Mn+2+, NO, ubiquinone; depression of antioxidant defence - reduced production of total
ceruloplasmin
, elevated blood levels of oxidized
ceruloplasmin
and enhanced catalase activity. It is suggested that malignant tumor growth is associated with marked paraneoplastic shifts in tissue redox-potential. These alterations are involved in mechanisms of paraneoplastic changes of red cells, microhemocirculation and circulation intensity. All these interrelated processes result in generalized paraneoplastic hypoxia of the organs and tissues. Basing on our and literature data, we propose an original scheme of the mechanisms of paraneoplastic disorders of tissue redox-status, microcirculation and erythrocytes.
