The effects of oral zinc supplementation on lipid peroxidation and the antioxidant defense system of alloxan (80-90 mg/kg)-induced diabetic rabbits were examined. Forty-five New Zealand male rabbits, 1 year old, weighing approximately 2.5 kg, were allocated randomly and equally as control, diabetic, and zinc-supplemented diabetic groups. After diabetes was induced, zinc-supplemented diabetic rabbits had 150 mg/L of zinc as zinc sulfate (ZnSO(4)) in their drinking tap water for 3 months. The feed and water consumption was higher in diabetic groups than (P<0.01) healthy rabbits. The body weight was lower in diabetic rabbits compared to control. The blood glucose levels were higher in diabetic groups than controls. The elevated plasma malondialdehyde (MDA) levels were determined in the diabetic group (P<0.01). The glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD), glutathione (GSH), and ceruloplasmin levels in the diabetic group were decreased by the effect of diabetes but there was no difference between zinc-supplemented diabetic and control rabbits. Serum zinc concentrations were lower in diabetic rabbits but iron (Fe) and copper (Cu) levels in sera were not different among the groups. As a result, it was concluded that daily zinc supplementation could reduce the harmful effects of oxidative stress in diabetics.

The present studies were designed to evaluate supplemental grape seed extract (GSE) and vitamin E supplements on lipid peroxidation, on antioxidant systems and peripheral blood lymphocytes in rats exposed to x-rays. Three groups of rats were investigated: a control group (CG) received intraperitoneal (i.p.) physiological serum 1 mL/day (n=10), i.p.; a vitamin E group (VG) received 50 mg/kg/day (n=10); an i.p. grape seed extract group received 50 mg/kg/day (n=10). Four weeks later, a 6 Gy radiation dose was given to the rats. Blood samples were taken 24 h later after irradiation and lymphocyte, malondialdehyde (MDA), reduced glutathione (GSH), nitrate, nitrite, reduced ascorbic acid, retinol, beta-carotene and ceruloplasmin concentrations were analysed. The levels of GSH (p<0.05), retinol (p<0.001), beta-carotene (p<0.05) and ceruloplasmin concentration (p<0.001) in the GSE group were found to be higher than in the control group but the level of MDA (p<0.001) and nitrite concentration (p<0.05) in rats supplemented with GSE were found to be lower than in the control group. The results indicate that GSE enhanced the antioxidant status and decreased the incidence of free radical-induced lipid peroxidation in blood samples of rats exposed to x-radiation. The antioxidant effect of GSE given to animals was more effective than vitamin E administered before whole-body irradiation in rats.

Previous studies revealed that oxidative stress could be an important component of the mechanism of organophosphate (OP) compound toxicity. The aim of the present study was to investigate both prophylactic and therapeutic effects of melatonin against fenthion-induced oxidative stress in rats. Therefore, we determined the changes in the levels of reduced glutathione (GSH) and malondialdehyde (MDA) in the whole blood, brain, pectoral muscle, liver, lung, heart, kidney, pancreas, and jejunum. Also, the changes in the levels of serum nitrite and nitrate, ascorbic acid, retinal, b-carotene, and ceruloplasmin were measured. In addition, activities of enzymatic antioxidants superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) in erythrocyte of normal and experimental animals were measured. It was found that fenthion administration increased the levels of MDA in all tissues and decreased or increased the levels of GSH in some tissues. In comparison to nitrate, nitrite and ascorbic acid levels in the serum of experimental groups, there was no significant difference between groups. However, fenthion toxicity led to decrease in retinol and beta-carotene levels; melatonin administration significantly prevented this decrease. Serum ceruloplasmin level was increased due to fenthion administration, but prophylactic and therapeutic melatonin administration inhibited the increase in ceruloplasmin level of serum. There was no significant change in SOD levels in melatonin-administered groups. Melatonin modulates the fenthion-induced changes in the activities of GPx and CAT. In conclusion, the results of the current study revealed that OP toxicity, induced by fenthion, activated oxidant systems in all antioxidant systems in some tissues. Melatonin administration led to a marked increase in antioxidant activity and inhibited lipid peroxidation in most of tissues.

1. The influence of dietary conjugated linoleic acid (CLA) on the antioxidant status in the absence or presence of endotoxin exposure was studied with male broiler chicks. 2. In experiment 1, a total of 240 1-d-old broilers were allotted into 4 dietary groups (0, 2.5, 5.0 or 10.0 g pure CLA/kg) to study the influence of CLA on growth performance and antioxidant defence systems. The results showed that growth performance was not altered by 42 d of CLA consumption. Increased total superoxide dismutase (TSOD) activities in liver, serum and muscle were observed in chicks given 10.0 g CLA/kg diet. Dietary CLA at 10.0 g/kg also markedly elevated liver catalase (CAT) activity. Malondialdehyde (MDA), a marker of lipid peroxidation, decreased in liver, serum and muscle in chicks given 5.0 and 10.0 g CLA/kg diet. 3. In experiment 2, a total of 120 1-d-old broilers were fed on a control diet (without CLA) or 10.0 g CLA/kg diet. Half of the birds fed on each diet were injected intraperitoneally with 0.25 mg/kg body weight of Salmonella enteritidis lipopolysaccharide (LPS) at 16, 18 and 20 d of age. Decreased glutathione peroxidase (GSH-Px), TSOD activity and increased ceruloplasmin and MDA concentrations were seen in the challenged chicks. Dietary CLA prevented the loss of body weight gain and feed conversion ratio of chicks followed repeated endotoxin exposure. CLA partially inhibited the increase of serum ceruloplasmin and MDA at 17 and 21 d of age and notably suppressed the decrease of serum TSOD activity at 21 d of age. 4. These results suggested that dietary CLA enhances the activity of antioxidant enzymes including TSOD and CAT. Supplementation of CLA has been shown to ameliorate the antioxidant balance and performance of chicks during oxidative stress.

Increased free radical activity in gestational diabetes (GDM) can lead to a host of damaging and degenerative maternal and fetal complications. Hence antioxidant levels in blood of GDM mothers and cord blood were estimated. Erythrocyte glutathione (GSH), superoxide dismutase (SOD) and thiobarbituric acid reactive substances (TBARS), plasma vitamins C and E and serum total glutathione-S-transferase (GST), protein thiols and ceruloplasmin (Cp) were estimated spectrophotometrically in maternal blood of age matched controls and mothers with GDM and also in cord blood samples of the above. There was a significant increase in the erythrocytic GSH, serum total GST and protein thiols in GDM maternal blood when compared to controls whereas erythrocytic SOD exhibited a marked decrease in GDM cases. The changes in plasma vitamins C and E, Cp and erythrocytic TBARS in GDM were not significantly different from controls. Cord blood levels of protein thiols were also significantly increased in GDM. No significant changes were observed in the serum Cp and GST levels of the same. Hence, elevated glucose levels can induce oxidative stress in GDM mothers.

The present study was aimed to investigate the possible pancreatic tissue protective nature of D-Pinitol, a cyclitol present in soybean, against free radical-mediated oxidative stress in streptozotocin-induced diabetic rats by assaying the activity of pancreatic enzymatic antioxidants such as superoxide dismutase (SOD), catalase, glutathione peroxidase (GPx) and glutathione-S-transferase (GST) and the levels of plasma non-enzymatic antioxidants such as vitamin E, vitamin C, ceruloplasmin and reduced glutathione (GSH). To assess the extent of oxidative stress, the levels of lipid peroxidation (LPO) and hydroperoxides in both plasma and pancreatic tissues were also measured. A significant increase in the levels of both lipid peroxides and hydroperoxides with a concomitant decrease in antioxidant status was observed in the diabetic rats when compared to control rats. Oral administration of D-Pinitol (50 mg/kg b.w./day for 30 days), a major cyclitol present in soybean, ameliorates the free radical-mediated alterations to near normalcy. The pancreatic tissue protective nature of D-Pinitol was further evidenced by histological observations. The results were statistically comparable with glyclazide, a standard hypoglycemic drug. Thus, the results of the present study suggest that D-Pinitol protects the pancreatic tissue from free radical-mediated oxidative stress in addition to its antidiabetic property.

Ruta graveolens L. (Rutaceae) are traditionally used for the treatment of rheumatism, arthritis and other inflammatory conditions in the traditional medicine of India, were evaluated for their protective effect in acute and chronic models of inflammation. Carrageenan induced rat paw edema and adjuvant induced arthritis were employed as the experimental models of acute and chronic inflammation respectively. Isolated polyphenolic and alkaloid fraction (AFR) from Ruta graveolens and evaluated its anti inflammatory activity in carrageenan induced acute model. AFR with a dose 10 mg/kg showed higher anti inflammatory effect than polyphenols and standard drug diclofenec. AFR significantly decreased the paw edema in arthritic rats. TBARS, COX-2, 5-LOX and MPO level were decreased and the levels of antioxidant enzymes and GSH level were increased on treatment with AFR. The increment in CRP level and ceruloplasmin level observed in arthritic animals were also found to be significantly restored in AFR treated rats. The results demonstrated the potential beneficiary effect of isolated polyphenolic and alkaloid fraction of Ruta graveolens L. on acute and chronic models of inflammation in rats.

Glutathione (GSH) depletion is often detected in chronic pathological conditions like hepatitis C infection, alcohol consumption or xenobiotic assault with simultaneous reactive oxygen species (ROS) generation and hepatic iron overload. However, relation between GSH depletion and regulators of iron homeostasis is not clear so far. To determine that hepatic HepG2 cells were treated with GSH synthesis inhibitor butathione sulfoximine (BSO) and a dual regulation of ceruloplasmin (Cp) that involves in hepatic iron release was detected unlike other iron homeostasis regulators. BSO treatment that caused marginal GSH deficiency increased Cp synthesis due to increased transcription mediated by activator protein (AP)-1-binding site. In higher GSH deficiency (> 40 %) with increased ROS generation, Cp expression was decreased due to promotion of Cp mRNA decay mediated by 3'untranslated region (3'UTR) as found by transfecting chimera of chloramphenicol acetyl transferase (CAT) gene with Cp 3'UTR. RNA gel shift assay showed significant reduction in 3'UTR binding protein complex in similar condition. Decreased CAT expression and RNA-protein complex binding are reversed by pretreatment with antioxidant N-acetyl cysteine suggesting 3'UTR binding protein complex is redox-sensitive. This unique and opposite regulation of Cp provides a mechanism of hepatic iron-deposition during glutathione deficiency detected in chronic pathological conditions.

Chronic exposure of pancreatic beta-cells to supraphysiologic glucose causes adverse beta-cell dysfunction. Thus, the present study was aimed to investigate the hypothesis that oral administration of resveratrol attenuates hyperglycemia, proinflammatory cytokines and antioxidant competence and protects beta-cell ultrastructure in streptozotocin-nicotinamide-induced diabetic rats. Oral administration of resveratrol (5 mg/kg body weight) to diabetic rats for 30 days showed a significant decline in the levels of blood glucose, glycosylated hemoglobin (HbA1c), TNF-alpha, IL-1beta, IL-6, NF-kappaB p65 unit and nitric oxide (NO) with concomitant elevation in plasma insulin. Further, resveratrol treated diabetic rats elicited a notable attenuation in the levels of lipid peroxides, hydroperoxides and protein carbonyls in both plasma and pancreatic tissues. The diminished activities of pancreatic superoxide dismutase (SOD), catalase, glutathione peroxidase (GPx) and glutathione-S-transferase (GST) as well as the decreased levels of plasma ceruloplasmin, vitamin C, vitamin E and reduced glutathione (GSH) in diabetic rats were reverted to near normalcy by resveratrol administration. Based on histological and ultrastructural observations, it is first-time reported that the oral administration of resveratrol may effectively rescue beta-cells from oxidative damage without affecting their function and structural integrity. The results of the present investigation demonstrated that resveratrol exhibits significant antidiabetic potential by attenuating hyperglycemia, enhancing insulin secretion and antioxidant competence in pancreatic beta-cells of diabetic rats.

The state of enzymatic antioxidant system and thyroid status in the rat tissues under early postnatal overnutrition leading to shortening of life-span and calorie-restricted diet extending lifespan have been studied. It was found that the concentration of thyroxine, body weight and content of lipid hydroperoxides in the blood serum and liver post-mitochondrial fraction of rats had been reliably increased. At the same time the concentration of GSH and GSSG, superoxide dismutase, Se-dependent glutathione peroxidase (GP) activities in the liver and GP activity in the blood serum of experimental rats were reduced. The use of calorie-restricted diet for 2 months showed considerable decrease of thyroxine and triiodothyronine concentration, body weight and the content of lipid hydroperoxides in the blood serum of experimental rats compared with control. At the same time GP activity in the liver, Se-dependent GP activity and content of ceruloplasmin in blood of experimental rats were essentially enhanced. The obtained results allow to conclude that from all studied indices the activity of GSH-dependent antioxidant system (especially Se-dependent GP activity) and thyroid status revealed the most sensible changes in response to the used kind of nutrition. Unlike hypocaloric diet overnutrition results in a decrease of Se-dependent GP activity in the liver and blood of rats. That may be the most crucial moment in the maintenance of prooxidant-antioxidant balance of tissues.
