Ceruloplasmin was isolated from mice blood serum by means of ion exchange chromatography and gel filtration. Its content in blood was equal to 10-12 mg/100 ml, oxidase activity--9-12 U/mg of protein at E440, molecular mass--161.3 +/- 5.8 kDa. Properties of mice
ceruloplasmin
were studied as compared with the human and rat
glycoprotein
, isolated using the similar procedure.

Immunological profile of 42 men working under exposure to chlorine for a period from 2 to 38 years was analysed. Toxicological analyses of air samples which were carried out for 20 years showed similar chlorine concentration ranging from 0.21 to 1.05 mg/m3. The number of B, T, and non-B, non-T lymphocytes was determined as well as the C3c, C4 complement components and proteins of acute phase reaction: alpha 1-acid-
glycoprotein
, haptoglobin and
ceruloplasmin
in the serum. Chronic exposure to chlorine produced a stimulating effect on the immune system, which was manifested by elevated IgA, IgD and IgM levels, the concentration of acute phase reaction proteins and increased number of B lymphocytes. The observed phenomenon had no influence on the antibacterial and anticancer immunity of the workers.

In a sample group of 92 women undergoing prenatal echo-guided transabdominal amniocentesis between the 12th and 23rd week of pregnancy the Authors analysed amniotic fluid and maternal serum using the recently developed method of high resolution protein electrophoresis in order to identify the presence of particular proteins in the amniotic fluid which are pathognomonic for a number of maternofetal pathologies. The results obtained in normal and pathological pregnancies or in the case of twins showed a marked dispersion in amniotic fluid of total protein concentrations depending on the period of gestation; in addition, albumin, alpha 1-antitrypsin, alpha 2-
glycoprotein
acid, alpha 2-macroglobulin and beta 2-protein were also found. Plasma levels of prealbumin, albumin, alpha 1-
glycoprotein
acid and IgG were slightly reduced, whereas there was a marked increase in
ceruloplasmin
, transferrin and fibrinogen; C3 and haptoglobin levels were normal. It is therefore possible to ascertain that amniotic fluid proteins analysed by high-resolution 15-band electrophoresis did not vary qualitatively or quantitatively until the 23rd week of gestation and in those cases of twin or pathological pregnancies examined no anomalous band was found in the protein electrophoresis of maternal serum or amniotic fluid which might prove useful in prenatal diagnosis.

Due to the stress imposed by the process of bone marrow transplantation (BMT), we hypothesized that individuals receiving such a transplant underwent an acute phase response (APR). Circulating levels of C-reactive protein (CRP), haptoglobin (HAP), alpha-1 acid
glycoprotein
(AAG),
ceruloplasmin
(
CER
), zinc (Zn), copper (Cu), interleukin-6 (IL-6), albumin (ALB), and thyroxine-binding prealbumin (TBPA), were measured at baseline (Day -7), Day -4, Day 0 (Transplant Day), Day +2, +7, and weekly until day 28 in 14 adults receiving an autologous bone marrow transplant as Phase 1 treatment for various hematologic or solid tumor malignancies. Ten of 14 recipients survived, 9 of which had a significant increase in CRP (p = 0.012), HAP (p = 0.011), AAG (p = 0.002), and decrease in ALB (p = 0.002) and TBPA (p = 0.004) on Day +7, but not Day 0, after bone marrow reinfusion. These findings document the presence of an APR and suggest that the bone marrow transplant process (post reinfusion) initiates a stress response in the recipient.

An objective approach for monitoring the treatment of acute pulmonary exacerbation in cystic fibrosis was evaluated. Eleven biochemical markers of inflammation (erythrocyte sedimentation rate, neutrophil count, C-reactive protein, alpha-1 antitrypsin, haptoglobin,
ceruloplasmin
, fibronectin, alpha-1
glycoprotein
, alpha-2 macroglobulin, C3, granulocyte elastase and anti-Pseudomonas IgG) were measured in blood serum and plasma from 46 cystic fibrosis patients with chronic Pseudomonas aeruginosa colonization before and after treatment. The overall outcome in each patient was evaluated by means of a pondered sum of clinical, chest X-ray and lung function scores. Biochemical markers were related to the overall clinical improvement: haptoglobin,
ceruloplasmin
, fibronectin and alpha-1
glycoprotein
showed a good sensitivity (64-70%), specificity (60-70%) and positive predictive value (86-89%). Granulocyte elastase showed a similar sensitivity (67%) and positive predictive value (85%) but a lower specificity (33%). The negative predictive value was generally poor (32-39%). Our data suggest that the combined measurement of some markers of inflammation and of conventional clinical parameters, may help in evaluating the efficacy of anti-infective treatment in cystic fibrosis.

It has been reported that microheterogeneity (M-HT) of serum glycoproteins including transferrin is found in alcoholic liver disease (ALD). In the present study, M-HT of serum glycoproteins in ALD patients was analyzed using the Western blotting technique after isoelectric focusing. M-HT was found in serum alpha 1-antitrypsin, alpha 2-macroglobulin,
ceruloplasmin
, alpha 1-acid
glycoprotein
and hemopexin as well as transferrin, but not in serum prealbumin. M-HT disappeared following treatment with sialidase in one group of glycoproteins, but not in another group of glycoproteins. In hemopexin, M-HT was recognized only after treatment with sialidase. These results suggest that mechanisms of the appearance of M-HT of serum glycoproteins in ALD may differ. One mechanism is the interference of glycosylation of glycoproteins in the Golgi apparatus, and another is the decrease of asialo-protein receptors in hepatocytes.

Recently, a few reports have shown that severe depression may be associated with higher levels of positive acute phase proteins (APPs), such as haptoglobin (Hp), alpha 1-acid
glycoprotein
(alpha 1S) and lower levels of negative APPs (visceral proteins), such as albumin (Alb) and transferrin (Tf). In order to reassess whether depression is related to alterations in the expression of plasma APP concentrations, we measured in 84 normal controls and depressed inpatients positive APPs such as Hp, alpha 1-antitrypsin (alpha 1AT), hemopexin (Hpx),
ceruloplasmin
(Cp), complement component C3C and one visceral protein, i.e., retinol binding protein (RBP). We found increased plasma concentrations of Hp, alpha 1AT, and Cp in major depressed subjects as compared with healthy controls, with minor depressives exhibiting an intermediate position. RBP was significantly lower in minor and major depressives than in normal controls. The disorders in these proteins were rather sensitive (62%) for major depression, with a specificity equalling 96%. Our findings are compatible with the hypothesis that major depression may be accompanied by inflammatory changes with higher levels of positive APPs (i.e., alpha 1AT, Hp, Cp, alpha 1S) and lower levels of visceral proteins (i.e., RBP, Tf, Alb).

Inflammation, an acute phase stress, alters copper (Cu) metabolism, but effects on human Cu requirements are unknown. Cu supplementation (2 mg/day, 4 weeks) increased erythrocyte Cu-zinc (Zn) superoxide dismutase (SOD) activity levels in 18 of 23 rheumatoid arthritis (RA) patients receiving gold or methotrexate (mean increase 21%). SOD values were significantly lower in RA patients than in 47 age- and sex-matched controls before, but not after supplementation. Supplementation did not significantly affect
ceruloplasmin
(Cp) activity or protein concentrations in either group. However, RA subjects showed significantly lower Cp activity to protein ratios compared to controls, before and after supplementation. Cu supplementation did not affect acute phase status of RA patients as evidenced by unchanged serum alpha-1-acid
glycoprotein
levels. In conclusion, the effects of Cu supplementation on erythrocyte SOD activities suggested a trend toward marginal Cu status in RA patients.

Conformational properties of
ceruloplasmin
were studied immediately in blood serum of healthy volunteers, patients with tuberculosis, with pulmonary cancer, with pneumonia and with Wilson-Konovalov disease. The
glycoprotein
conformation was found to depend on the volunteer physiological state and/or available pathology. The
ceruloplasmin
conformation and status of its copper-containing sites of the I type affected the enzyme oxidase properties and hence routine colorimetric procedures require some corrections for estimation of
ceruloplasmin
concentration and activity in blood serum.

We determined serum levels of alpha 1-antitrypsin, alpha 1-acid
glycoprotein
,
ceruloplasmin
, haptoglobin, alpha 2-HS
glycoprotein
, C3 and fibronectin in 22 patients with acute pneumonia, 10 patients with mycoplasma pneumonia and 8 patients with exacerbation of chronic bronchitis at both acute and convalescent phases to clarify whether levels of these proteins change with inflammation. Serum levels of alpha 1-antitrypsin, alpha 1-acid
glycoprotein
,
ceruloplasmin
, haptoglobin and C3 in the patients in the acute phase were significantly higher than that in the patients in the convalescent phase. The serum level of fibronectin in the patients in the acute phase was significantly lower than that in the patients in convalescent phase. The serum level of alpha 2HS
glycoprotein
remained unchanged.
