A distinctive periadnexal multinodular granulomatous dermatitis of unknown cause was diagnosed over a 6-year period in 22 dogs (13 males and nine females) aged 8 months to 10 years, purebred as well as mixed-breed. Nodules, usually more than two, started on the face and were often unaccompanied by signs of irritation. All dogs were otherwise healthy except a Viszla with bilateral anterior uveitis and a Basset hound with nasal congestion and sneezing. Microscopically, cutaneous lesions occurred in a periadnexal pattern, replaced sebaceous glands, coalesced, and extended into the subcutis and in more advanced cases through the panniculus carnosus. Infectious agents were not demonstrated by aerobic, anaerobic, or fungus cultures or by light or electron microscopy, nor were ectoparasites seen. Immunofluorescence and peroxidase-antiperoxidase techniques gave negative results for intraepidermal and basement membrane deposits of IgA, IgG, IgM, and C3. Lesions were unresponsive to a variety of antibiotics; they regressed spontaneously in two untreated dogs and rapidly in dogs given only corticosteroid therapy. Continuous low-dose corticosteroids were necessary in a few dogs to prevent recurrence.

Methimazole has been reported to produce extensive degenerative changes in olfactory epithelium and a severe deficit in odor detection [Genter BM, Owens DM, Carlone HB, Crofton KM. Fundam. Appl. Toxicol. 1996;29:71-77; Genter BM, Owens DM, Deamer NJ, Blake BL, Wesley DS, Levi PE. Toxicol. Pathol. 1995;23:477-486.]. To examine this further, rats were tested on olfactory detection and discrimination problems before and after intraperitoneal injection of 300 mg/kg methimazole. In the first 2 days after treatment, experimental rats had nasal congestion and a modest decrement on odor detection and odor mixture discrimination tasks. They performed almost as well as control rats on the third post injection day. In a separate group of rats, anterograde transport of horseradish peroxidase from olfactory epithelium to the bulb was examined 1, 2, 3, and 5 days after administration of methimazole. The treatment produced a modest but progressive disruption of bulbar input: 2 days after administration only approximately 10% of bulbar glomeruli had reduced levels of reaction product while 30-40% of glomeruli had little or no reaction product in 3-5 day survival rats. These results indicate that methimazole is not a particularly effective olfactotoxin and does not produce anosmia or even a severe hyposmia.

In the present experiments, the possible role of curcumin in ovalbumin induced allergic rhinitis in guinea pig model was investigated. Various allergic rhinitis symptoms viz sneezing, rubbing frequencies, lacrimation and nasal congestion at various humidity conditions as well as on repeated sensitization were studied. The biochemical changes like serum IgE, IL-4 and nitric oxide (NO) in nasal lavage and eosinophil peroxidase activity in nasal homogenates were determined in allergic rhinitis. Curcumin treatment significantly reduced the symptoms (sneezing, rubbing frequencies, lacrimation and nasal congestion) and improved the histopathological alterations (reduction in inflammatory cells infiltration) of nasal mucosa in allergic rhinitis. Furthermore, curcumin treatment prevented significantly elevation of serum IgE, IL-4, NO in nasal lavage and eosinophil peroxidase in nasal homogenate. In the present experimental findings, we suggest that curcumin is a promising anti-allergic agent that may be useful in the clinical management of allergic rhinitis.
