To compare the efficiency of hybridization methods for the detection of HPV genome, 22 cases of invasive squamous cell carcinoma of the uterine cervix were analyzed by Southern blot analysis and in situ hybridization carried out with 35S- and biotin-labeled probes. These cases contained from less than one to as many as 50 copies per cell of HPV 16 and 18 types. To increase the sensitivity of biotinylated probes, a silver enhancement procedure of the peroxidase reaction product was applied. Results showed that in situ hybridization performed with isotopic probes is as sensitive as Southern blot analysis and is more sensitive than that performed with biotin-labeled probe. However, the application of the silver enhancement procedure increases the percentage of HPV-positive cases from 27 to 50%.

We investigated populations of the infiltrating cells in Bowen's disease (BD) and squamous cell carcinoma (SCC), both of which arose in the same patient, using the Avidin-Biotin-peroxidase complex method with eight monoclonal antibodies. T lymphocytes were most predominant among infiltrating cells; NK cells, B cells, and monocytes were rarely seen in either BD or SCC. Analysis of subsets of the infiltrating T lymphocytes revealed that the number of suppressor/cytotoxic (s/c) T cells was twice that of helper/inducer (h/i) T cells in BD, while the number of s/c T cells was lower than that of h/i T cells in SCC. The immunohistochemical results in the present case differed from those of predominant infiltration of h/i T cells and of s/c T cells in three other reports of BD and SCC. These results suggest that the population of the cellular infiltrates may be modulated by the nature of tumors and by the immuno-competent state of the hosts.

The expression of the ras p21 in oral squamous cell carcinoma was examined immunohistochemically with the use of a monoclonal antibody NCC-RAS-001 with the avidin-biotin-peroxidase complex method. The expression of ras p21 product was detected in 65.7% (44 of 67 cases) of cancer patients. On the basis of the degree of histologic differentiation of the cancer cells, the incidence of ras p21 was found to be as follows: 63.4% (26 of 41 cases) were well differentiated; 86.7% (13 of 15 cases) were moderately differentiated; and 45.5% (5 of 11 cases) were poorly differentiated. The highest incidence was found in patients in their sixties--80.0% (16 of 20 cases). The incidence decreased to 40% in patients over 80 years of age. The incidence of ras p21 on the basis of location was as follows: 81.8% (9 of 11 cases) involved the buccal region, 70.6% (12 of 17 cases) were in the gingiva, and 55.0% (11 of 20 cases) were in the tongue.

A 62-year old male was admitted to our hospital because of fever and dysphagia on November 14, 1987. The peripheral leukocyte count was 174,400/microliters with 93% blasts and bone marrow aspiration showed that 90.4% of nucleated cells were blasts positive for both myeloperoxidase and alpha-naphthylbutyrate esterase. Chromosome analysis revealed a karyotype of 45XY, 9q+, 16q+, -20 and 22q-. Esophageal X-ray and endoscopy showed abnormalities. Esophageal biopsy revealed squamous cell carcinoma. He was diagnosed as having Ph1 positive acute myelomonocytic leukemia (AMMoL, M4) and esophageal cancer. He was treated with BHAC-DMP and intermediate-dose ara-C therapy for leukemia and a complete remission was obtained by March 25, 1988. As treatment for esophageal cancer, radiation therapy (total 4,200 cGy) was given and followed by chemotherapy with CDDP and 5-FU. However he died on April 8, 1988. Autopsy findings showed disseminated invasion of esophageal cancer. Ph1 positive AMMoL associated with esophageal cancer is extremely rare.

A case of urothelial tumor with extremely high serum carcinoembryonic antigen (CEA) levels is described. A 68-year-old female presented with macroscopic hematuria and left flank pain. Laboratory examination revealed an extremely high serum level of CEA (194 ng/ml) and elevated levels of serum CA 19-9 (235 U/ml) and squamous cell carcinoma (SCC)-Antigen (10.7 ng/ml), while urine CEA remained within normal limits. No abnormal findings were recognized in gastrointestinal and respiratory systems, but left renal pelvic tumor (T4N2M0) was discovered. Nephroureterectomy with regional lymph node dissection was done. The pathologic anatomy was infiltrating non-papillary transitional cell carcinoma (TCC, G2 = G3, pT4N2M0). More than 30% of the tumor cells were positive for CEA by ABC-peroxidase staining. Levels of tumor markers remained higher than normal after the operation and were normalized after M-VAC (methotrexate, vinblastine, adriamycin and cisplatin) chemotherapy. However, 6 months after the operation, levels of tumor markers rose again and lung metastases appeared. She died 10 months after the operation.

Forty-seven cases of oral squamous cell carcinoma were examined immunohistochemically by the avidin-biotin peroxidase complex method with anti-epidermal growth factor (EGF) receptor mouse monoclonal antibody, and 24 cases (51%) were shown to have EGF receptor-positive cells. Correlation was strong between presence of EGF receptors and differentiation; the EGF receptor-positive cells were differentiated, whereas poorly differentiated tumors exhibited less intense staining. EGF receptor gene and c-erb B-1 by Southern blot analysis disclosed that one of 25 cases of squamous cell carcinoma exhibited a fourfold amplification of the gene.

Because the sensitivities of individual hybridization techniques differ considerably, their role in accounting for the published frequencies of human papillomavirus (HPV) DNA in anal squamous cell carcinomas, ranging from 0 to 61%, must be investigated. With the use of biotinylated probes to HPV 6, 11, 16, 18, and 33, three hybridization techniques were performed on the same paraffin-embedded tissue blocks selected from 13 cases of anal squamous cell carcinoma. HPV DNA was detected in 0%, 62%, and 85% of cases with the use of in situ hybridization with horseradish peroxidase, in situ hybridization with alkaline phosphatase, and dot blot hybridization, respectively. By dot blot hybridization, 69% had HPV 16/6 and 15% had HPV 6/11. An HPV DNA frequency range of 0-85% in the same group of tumors with the use of three hybridization techniques indicates the influential role of the method on HPV DNA prevalences. HPV DNA was identified regardless of patient gender or type of squamous cell carcinoma. The presence of HPV 16 in 82% of the positive cases in supportive evidence of the carcinogenic role of the HPV in anal squamous cell carcinoma.

To evaluate the clinical significance of monoclonal antibody against human pulmonary surfactant apoprotein (S-AP), surgically resected lung cancer from 122 patients was studied. Paraffin embedded tissues were used for the immunohistochemical study by the avidin-biotin-peroxidase complex method. The results were as follows. 1. Adenocarcinoma showed highest immunoreactivity for S-AP compared to the other histologic types. Among subtypes of adenocarcinoma, type II alveolar epithelial type, clara cell type and mixed type of these two types were strongly positive (100%, 77.8% and 66.7% respectively). These results indicate that this antibody may be a good marker for the subtyping of adenocarcinoma. 2. There were some positive cases in other histologic types especially in peripheral type of squamous cell carcinoma. These findings suggest that this antibody was useful for the histological differentiation of lung cancer. 3. As to the immunohistochemical reactivity there was a good correlation between tissue and cytological specimens, which indicate cytological studies may be adequate for this kind of histopathological studies. 4. In our study, there were no patients with S-AP positive carcinomas other than patients with lung cancer. These results indicate that this antibody could be used for the differential diagnosis between primary and secondary lung cancer.

The expression of a unique beta-tubulin isoform (class III) was monitored in squamous cell carcinoma (SCC) and normal epithelial cells using a monoclonal tubulin antibody called TuJ1. Whole tissue homogenates of SCC, normal tissue, SCC grown in nude mice, and SCC cultured cells were examined using sodium dodecyl sulfate polyacrylamide gel electrophoresis and Western blot. TuJ1 antibody localization was performed using peroxidase immunostaining on paraffin sections of SCC, normal tissue, nude mouse SCC, and immunofluorescent microscopy of SCC cultured cells. The malignant tissues examined stained positive with TuJ1 and a general beta-tubulin antibody, whereas the normal tissues stained positively only for the general beta-tubulin antibody. TuJ1 epitope expression may be a useful marker for SCCs and may assist in understanding differences between normal and malignant squamous cells.

Twenty patients with bladder carcinoma (14 transitional cell carcinoma and six squamous cell carcinoma) and ten controls of (four with bilharzial lesions, three without and three with metaplasia and dysplasia) were subjected to immunocytochemical and immunohistochemical studies. Bladder and urine samples examination were carried out with the high molecular weight cytokeratin and broad spectrum keratin antibodies using peroxidase anti-peroxidase. Cytokeratins of high molecular weight were expressed in all squamous cancer while broad spectrum keratin was positive in 83.3%. The transitional cell carcinoma was positive in 50% with both cytokeratin and broad spectrum keratin. Urine shedded were positive in 83.3% and 66.7% of squamous cell carcinoma with high molecular weight cytokeratin and broad spectrum keratin respectively. However, each of them was positive in 50% of transitional carcinoma, and negative with non-malignant cases except with metaplasia and dysplasia.
