We have investigated transformation of eight industrial dyes by a white-rot fungus, Trametes versicolor. The fungus was found to decolorize Reactive Golden Yellow R, Procion Red, Reactive Violet 5, Reactive Blue 28, and Ponceau Red 4R at an initial dye concentration of 80 ppm within 72 h of incubation, whereas it took 5 d to completely decolorize Reactive Black 5 (40 ppm). However, it did not significantly decolorize Reactive Red 152 and Novatic Blue BC S/D. During decolorization in liquid medium, laccase and manganese-independent peroxidase (MiP) activities were detected in culture filtrate of T. versicolor. Dye-decolorizing activity of the culture was found to be associated with H2O2-dependent activity of the culture filtrate. Furthermore, dye-decolorizing activity of the culture filtrate was not influenced by Mn2+ or veratryl alcohol, thus suggesting a role of extracellular MiP in decolorization of synthetic dyes by T. versicolor.

Stereum hirsutum, a white rot fungus, has a good growth in solid state fermentation. This was carried on with wheat bran, soy bran and a mixture of both. Mycelia grown on soy bran showed the highest decolorization activity on Ponceau 2R (xylidine), indigo carmine and malachite green. Optimal relationship between decolorization and detoxification of malachite green was 30 g of fresh weight (mycelia plus substrate) in 500 ml malachite green solution, 42 U/l of laccase was measured in this solution. Decolorization was carried on without the addition either of nutrients or mediators. Conditions for maximal decolorization did not agree with those for maximal ligninolytic enzyme production, but effectiveness of laccase activity on decolorization was evidenced by electrophoretic analysis, that allowed laccase identification and its decolorization activity in gels stained with indigo carmine and malachite green, with ABTS as mediator. Detoxification was assayed using the sensible fungus Phanerochaete chrysosporium.

In this paper the efficiency of the combined action of laccase and cellobiose dehydrogenase (CDH) to decolourise different synthetic dyes such as Remazol Brilliant Blue R (RBBR), Methyl Green (MG), Direct Violet (DV), Ponceau Xylidine (PX), Bismark Brown (BB) and Poly R-478 (PR) was assessed. It was found that the use of CDH could be a promising alternative to the utilisation of the expensive and poisonous chemical mediators such as HOBT although much research on this topic remains still to be done.
