In the present study, a pot culture experiment was conducted to estimate the ameliorating effect of propiconazole (PCZ) on drought stress in cowpea (Vigna unguiculata (L.) Walp.) plants. From 30 days after sowing (DAS), the plants were subjected to 3, 6 and 9 days interval drought (DID) stress and drought stress with PCZ at 15 and 15 mg l(-1) PCZ alone and 1 day interval irrigation was kept as control. The plant samples were collected on 34 DAS (3 DID), 37 DAS (6 DID) and 40 DAS (9 DID). The plants were separated into root, stem and leaf for estimating the antioxidant contents and activities of antioxidant enzymes. Individual and combined drought stress and PCZ treatments increased ascorbic acid (AA), alpha-tocopherol (alpha-toc) contents, superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT) and polyphenol oxidase (PPO) activities when compared to control. The PCZ treatment mitigated the adverse effects of drought stress by increasing the antioxidant potentials and thereby paved the way for overcoming drought stress in V. unguiculata plants.

A pot culture experiment was conducted to estimate the drought stress mitigating effect of ketoconazole (KCZ), a fungicide cum plant growth regulator, in Catharanthus roseus plants. The plants under pot culture were subjected to drought stress and drought stress with KCZ from 30 days after sowing (DAS) and regular irrigation was kept as control. Antioxidant contents and activities of antioxidant enzymes were estimated from root, stem and leaf of both control and treated plants. The alkaloid ajmalicine was extracted and estimated from the roots of control, drought stressed and KCZ treated plants. Individual and combined drought stress and KCZ treatments increased ascorbic acid, alpha-tocopherol contents, superoxide dismutase, ascorbate peroxidase, catalase and polyphenol oxidase activities when compared to control. There was a significant enhancement in ajmalicine production under KCZ treated plants under drought stress when compared to well watered control as well as drought stressed plants. The KCZ treatment resulted in partial mitigation of drought stress by increasing the antioxidant potentials in C. roseus plants.

The effect of paclobutrazol (PBZ) treatments on the antioxidant metabolism of white yam (Dioscorea rotundata Poir.) was investigated in the present study. PBZ @ 15 mg l(-1) plant(-1) was given to plants by soil drenching, 30, 60, and 90 days after planting (DAP). The non-enzymatic antioxidant contents like ascorbic acid (AA), reduced glutathione (GSH) and alpha-tocopherol (alpha-toc), activities of antioxidant enzymes like superoxide dismutase (SOD), ascorbate peroxidase (APX), polyphenol oxidase (PPO) and catalase (CAT) were extracted and assayed on 100 DAP from leaf, stem and tubers of both control and PBZ treated plants. It was found that PBZ has a profound effect on the antioxidant metabolism and caused an enhancement in both non-enzymatic and enzymatic antioxidant potentials under treatments in white yam. Our results have good significance, as this increase the innate antioxidant potential of this food crop, which is helpful to satisfy the needs of antioxidants in diet and thereby make it an economically important food crop.

The variations in antioxidant potentials and indole alkaloid content were studied in the present investigation, in two varieties (rosea and alba) of Catharanthus roseus, an important herb used in traditional as well as modern medicine, exposed to water deficit stress. The antioxidant and alkaloid profiles were estimated from root, stem, leaf, flowers and pods. The antioxidant potentials were examined in terms of non-enzymatic antioxidant molecules and activities of antioxidant enzymes. The non-enzymatic antioxidant molecules studied were ascorbic acid (AA), alpha-tocopherol (alpha-toc) and reduced glutathione (GSH). The estimated antioxidant enzymes were superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), peroxidase (POX) and polyphenol oxidase (PPO). The antioxidant concentrations and activities of antioxidant enzymes were high under water deficit stress in all parts of the plants. Indole alkaloid content was high in the roots of rosea variety in response to stress when compared to alba variety.

The significance of plant growth-promoting rhizobacteria (PGPR) mediated increase in antioxidant potential in vegetables is yet unknown. The plant growth-promoting bacterium Bacillus lentimorbus NRRL B-30488 (B-30488) mediated induction of dietary antioxidant in vegetables ( Trigonella foenum-graecum, Lactuca sativa, Spinacia oleracea, and Daucus carota) and fruit ( Citrus sinensis) after minimal processing (fresh, boiled, and frozen) was tested by estimating the total phenol content, level of antioxidant enzymes, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide scavenging activities along with integral radical scavenging capacity by photochemiluminescence assay and inhibition of lipid peroxidation. Minimal processing of vegetables showed that T. foenum-graecum had the highest phenol content in B-30488-treated plants followed by L. sativa, D. carota, and S. oleracea. Thermally treated vegetables T. foenum-graecum (26-114.5 GAE microg mg (-1)) had an exceptionally high total phenolic content, followed by D. carota (25.27-101.32 GAE microg mg (-1)), L. sativa (23.22-101.10 GAE microg mg (-1)), and S. oleracea (21.87-87.57 GAE microg mg (-1)). Among the vegetables and fruit used in this study for enzymatic estimation, induction of antioxidant enzymes, namely, polyphenol oxidase (PPO), ascorbate peroxidase (APX), catalase (CAT), and superoxidase dismutase (SOD), was observed in edible parts of T. foenum-graecum, L. sativa, S. oleracea, and D. carota, after inoculation with B-30488. The scavenging capacity of the vegetables treated with B-30488 against DPPH and superoxide anion radical activity was found to be significantly high as compared to nontreated control. Mild food processing had no adverse effect on radical scavenging capacity. Photochemiluminescence also ascertains the above findings. The ability of the plant extracts to protect against lipid peroxidation and its ability to prevent oxidation of reduced glutathione (GSH) was measured in rat liver homogenate, and the results suggested that the inoculated plant exhibited better activity in all of the screened plants. Significant increases in shoot length, root length, and dry weight, averaging 164, 132, and 135% in T. foenum-graecum, 174, 141, and 156% in L. sativa, 129, 141, and 59%, in S. oleracea, and 125, 146, and 42% in D. carota, respectively, over untreated controls, were attained in greenhouse trials. To the best of the authors' knowledge, this is the first report of PGPR-mediated induction of antioxidant enzyme activity (PPO, APX, CAT, and SOD) along with the antioxidant activity of the extracts in both in vitro (DPPH radical scavenging and superoxide scavenging) and ex vivo conditions using the rat liver tissue (percent inhibition of lipid peroxidation and prevention of oxidation of GSH) and phenolic content. The results demonstrate the PGPR-mediated induction of antioxidant level in vegetables and fruit controls oxidative damage even after minimal processing and thus is indicative of its potential as a viable substitute of synthetic antioxidants.

A pot-culture experiment was conducted to estimate the ameliorating effect of triadimefon (TDM) on drought stress in sunflower (Helianthus annuus L.) plants. The plants were subjected to 3-, 6-, and 9-day-interval drought (DID) stress and drought stress with TDM @ 15 mg l(-1) and 15 mg l(-1) TDM alone from the 30th day after sowing (DAS). One-day-interval irrigation was kept as control. The plant samples were collected on and separated into root, stem and leaf for estimating the amino acid (AA), proline (PRO) and glycine betaine (GB) contents and the activities of antioxidant enzymes. Individual and combined drought stress and TDM treatments increased AA, PRO and GB contents, superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), and polyphenol oxidase (PPO) activities when compared to control. From the results of this investigation, it can be concluded that the application of TDM caused a partial amelioration of the adverse effects of drought stress by its influence on quaternary ammonium compounds and antioxidant potentials in H. annuus plants.

The objective of this study was to investigate the effects of different concentration (0, 0.1, 0.7, 1.5, 3, 6 and 9 mM) of SA on antioxidant enzymes in Capsicum annuum L. plants. Enzyme activities of peroxidase, polyphenol oxidase, ascorbate peroxidase, catalase and glutathione reductase were measured. The plants were grown in pots vermiculite. Before applying the salicylic acid treatments, plants filled with were irrigated with based nutrient solution (Hoagland solution) for 5 weeks. After 5 weeks, plants were sprayed with different concentrations (0, 0.1, 1.5, 3, 6 and 9 mM) of salicylic acid. Data were analyzed using SPSS software and means were compared by Duncan test. In each experiment 4 replicats were used. Concentrations of 1.5, 3, 6 and 9 mM of SA caused significant increase in polyphenol oxidase and peroxidase enzyme activities in treated leaves. Concentrations of 0.7, 1.5 ad 3 mM of SA decreased in (catalase, ascorbate peroxidase and glutathione reductase) activities, but concentrations of 6 and 9 Mm of SA increased enzyme activities. Different concentrations of salicylic acid had different effects on enzyme activities in Capsicum annuum L.

In the present investigation, Strychnos nux-vomica, an important plant used in traditional medicine, was evaluated for its antioxidant potential. The antioxidant potentials were examined in terms of non-enzymatic antioxidant molecules and activities of antioxidant enzymes. The non-enzymatic antioxidant molecules studied were ascorbic acid, alpha-tocopherol and reduced glutathione. The estimated antioxidant enzymes were superoxide dismutase, ascorbate peroxidase, catalase, peroxidase and polyphenol oxidase. The analyses were carried out in the field-collected leaf samples. It was found that plant contained a significant quantity of non-enzymatic and enzymatic antioxidants in the leaves. These findings have high significance in the pharmacological industry; however, a detailed investigation is needed to confirm this conclusion, by radical scavenging ability, which is underway in our laboratory.

Field-grown olive trees (Olea europaea L. cv. Chemlali) were used over two growing seasons to determine the effects of different saline water irrigation levels on levels of proline and chlorophyll contents and activities of superoxide dismutase (SOD), polyphenol oxidase (PPO), ascorbate peroxidase (APX), and catalase (CAT). The plants were irrigated with fresh water (FW; ECe = 1.2 dS m(-1)) and saline water (SW; ECe = 7.5 dS m(-1)). Leaf water relations (relative water content, water potential), photosynthetic activity, and leaf chlorophyll content decreased under irrigation with saline water. In spring 2005, net photosynthesis of young leaves was 24.5 and 14.9 micromol m(-2) s(-1) in FW- and SW-treated plants, respectively. In old leaves, these rates were 20.2 and 12.2 micromol m(-2) s(-1), respectively. The relative reduction of net photosynthesis in SW-treated plants varied from 39 to 46% and from 39 to 61%, compared to FW-treated plants during the first and second crop seasons, respectively. The relative reduction of leaf chlorophyll (a + b) content under high water salinity level exceeds 50%, compared to FW-treated plants. However, proline content and activities of SOD, CAT, and APX increased under saline water irrigation. The increase of proline content was more important in leaves than in roots. In young leaves, the increment of antioxidant activities in SW-treated plants was 2.67, 3.61, and 1.85 times, respectively, for SOD, APX, and CAT, compared to FW-treated plants. From these results, interaction between antioxidant defense system and proline contents seems to be involved in the salt tolerance mechanisms of Chemlali olive tree.

The ability of exogenous compatible solutes, such as proline, to counteract salt inhibitory effects in olive plants ( Olea europaea L. cv. Chemlali) was investigated. Two-year-old olive trees were subjected to different saline water irrigation levels supplied or not with exogenous proline. Leaf water relations (relative water content, water potential), photosynthetic activity, and leaf chlorophyll content decreased under either saline water level. The proline supplement mitigated the reduction of growth and photosynthetic activity under salt stress, and the mitigating effect of proline was different among treatments. The increment rate of leaf relative water content (RWC) in the presence of 25 and 50 mM proline was 4.45 and 6.67%, respectively, in comparison to values recorded in SS1-treated plants (plants irrigated with water containing 100 mM NaCl). In SS2 (200 mM NaCl) plus proline-treated plants, this increase was 1.14 times for 25 mM proline and 1.19 times for 50 mM proline higher than those recorded in severe salt stress treatment (SS2). In response to salt stress, Chemlali olive plants seem to activate a complex antioxidative defense system that was displayed via the increase of activities of superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) and the decrease of polyphenol oxidase (PPO) under either salt stress treatment. The exogenous application of proline improved the antioxidative enzyme activities of salt-stressed olive plants. Indeed, in young or old leaf tissues, the highest levels of these antioxidant enzymes activities were recorded in (SS2 + P2)-treated plants (plants irrigated with water containing 200 mM NaCl plus 50 mM proline). In young leaves, this increase was 2.11, 2.96, and 2.76 times, respectively, for SOD, APX, and CAT enzyme activities in comparison to their respective activities in control plants (nonstressed plants irrigated with fresh water). In old leaves, this increase was 2, 2.41, and 2.48 times, respectively, for the various enzymes. If compared to high water salinity-treated plants (SS2), this increase was 1.1, 1.3, and 1.4 times in young leaves, respectively, for SOD, APX, and CAT activities. From these results, the proline supplements seem to improve olive salt tolerance by amelioration of some antioxidative enzyme activities, photosynthetic activity, and, so, plant growth and the preservation of a suitable plant water status under salinity conditions. More to the point, the decrease of soluble sugars contents in proline treated-plants revealed the important osmoprotectant effect played by the added proline in such a way that limited the need of salt-stressed plants for soluble sugars synthesis.
