Genetic variation at 26 loci in a natural population of Anopheles aquasalis has been studied by zymogram technique. Average proportion of polymorphic loci is 23% (criterion I) and 34% (criterion II). On the average 8.1% of the genome is in heterozygous condition in each individual. The degree of genetic variability varies from locus to locus. Some enzymes such as AO, alpha-GPDH, 3 loci of HK, 3 loci of LAP, 3 loci of
MDH
, GOT, 3 loci of ODH and one locus of
EST

-4), are monomorphic. Of the rest, proportion of heterozygosity varies from the minimum of 0.041 at Xdh-2 to a maximum of 0.493 at Est-2. Similarly, except for the loci Est-2 and Est-3, proportion of heterozygote individuals is extremely low. A. aquasalis has a higher genetic identity with A. evansae (I = 0.625) than with A. argyritarsis (I = 0.543). There seems to be direct correlation between the genetic variability of a species and its capacity to explore diverse ecological nitches.

Using a database of allozyme studies, correlations in heterozygosity between selected enzyme loci (

, alpha GPDH, IDH, 6PGDH, LDH, SOD, AAT, PGM,
EST
, PGI) were calculated across vertebrate species. Large and positive correlations were observed with untransformed heterozygosity values. However, after transformation to correct for mean species heterozygosity, correlations were substantially reduced and median values were closer to zero. Some enzymes were more often involved in significant correlations than others, and correlations calculated across species within vertebrate classes were significant for different enzyme pairs in different classes. There was no evidence that significant correlations occurred primarily between functionally related enzymes. It is suggested that the observed correlations are best explained by variation between enzyme loci in functional constraint and effective neutral mutation rate.

Spontaneous mutation rates at ten allozyme loci on chromosomes II and III of Drosophila melanogaster were studied. Over the three and a half years study, one alpha-GPD mutation and two different IDH mutations were obtained. The alpha-GPD mutation was inherited in the Mendelian fashion, as expected. The two IDH mutations were peculiar in that the band of new types appeared only in females. In males, only the original bands were stained, and the positions where mutant alleles' bands should be present were blank. Both IDH mutant homozygotes appeared as null allele homozygotes, while in females clear-cut single bands were present.-The rates of spontaneous mutation varied greatly. Eight loci studied (

, MPI, NADD, OCT, PGM, 6PGD, SDH, SOD, XDH) analyzed. 40 loci and 117 alleles were detected in the Oncomelania. Both of GPI and PGM-I, with 7 alleles, were the most variable loci. 22 loci had more than 3 alleles each. Of 40 loci examined in the 24 isozyme systems, 14 were found to be polymorphic, the proportion of multilocus enzymes being 58.3%. Sixty strains of Bacillus sphaericus, including 31 insect pathogens were studied by multilocus enzyme electrophoresis and were classified into 44 zymovars (electrophoretic types). Among the entomopathogenic strains, 11 belong to the same zymovar (Z59) indicating a widespread frequent genotype. Bands of enzyme activity were not detected among the strains for the loci GPI (E.C.5.3.1.9), G6P (E.C.1.1.1.49), 6PG (E.C.1.1.1.44) and ME (E.C.1.1.1.40). The enzymatic loci NP (E.C.2.4.2.1) and ACON (E.C.4.2.1.3) were monomorphic while the other enzymes,

The genetic variation in the non-insect pathogenic group seemed to be greater than in the entomopathogenic group. This latter group appears to be distinct from other strains of these species. All insect pathogens were recovered in the same phenetic cluster and a diagnostic allele is reported for the identification of entomopathogenic strains.

associated with highly purified chloroplasts isolated from Arabidopsis and spinach. A cDNA clone encoding the novel enzyme was found in the Arabidopsis

NAD-dependent malate dehydrogenase
. A derived amino acid sequence is very similar to mitochondrial and peroxisomal NAD-MDHs within the region coding for the mature protein but possesses a 80-amino acid long N-terminal domain with typical characteristics of a chloroplast transit peptide. In vitro synthesized labeled precursor protein was imported into the stroma of spinach chloroplasts and processed to a mature enzyme subunit of 34 kDa. Expressed in Escherichia coli, the recombinant enzyme exhibited the same distinctive isoelectric point of 5.35 as the original enzyme from Arabidopsis chloroplasts. Northern analysis revealed that the protein is expressed in both autotrophic and heterotrophic tissues. The findings reported here indicate that the "malate valve" operates not only in the illuminated chloroplasts but also in dark chloroplasts and in heterotrophic plastids and is therefore a general mechanism to maintain the optimal ratio between ATP and reducing equivalents in plastids.

Isozyme variation among 54 isolates of Microsporum canis, 18 Microsporum cookei isolates and two Diheterospora isolates were studied using starch gel electrophoresis. Of eight enzymes examined, four were polymorphic (

, G6P,

and PEP), having from two to four electrophoretic forms. Within each species, consistent and reproducible isozyme patterns of the eight enzyme systems were obtained. Phenotypic diversity (H) in M. canis was higher than in M. cookei (H = 0.459 and H = 0.408 respectively), but phenotypic differentiation of M. canis isolates from different geographical regions (Auckland, Wellington and Palmerston North, New Zealand) was low, with a proportion of total diversity (Gst) of 0.151 found among the localities. The results suggest that the isolates of M. canis from different geographical regions are closely related, supporting the theory of a common lineage.

A special type of variation--the mixoploid variation, which had a great effect on the variation in the production of banana vitroplants, was reported for the first time. By means of examining chromosome, isozymes (

, POX) and endogenous hormones (iPAs, IAA), the varied characters of the mixoploid varianties during the propagation in vitro when cultured as explant were investigated. The rate of the chromosome aberration in cells was characterized by starting at a much higher level and rising more rapidly in the preceding 15 subcultures as compared with the control. The rate of the isozyme variation was higher obviously than that of the control. The plantlets cloned in vitro from the mixoploid variant were also studied. The experiment showed that both the variant rates of chromosome and phenotype rose obviously. The mixoploid variant can cause the rate of variation to rise during the micropropagation.
