Although the human and the nonhuman primate fetal adrenal glands undergo a highly unique pattern of cortical zone-specific intrauterine growth and development, studies of the regulatory components of the cell cycle responsible for this growth have not been conducted. Therefore, the present study determined expression of the cell cycle regulators, cyclin D1 and cyclin E, and their cyclin-dependent kinases, Cdk2, Cdk4, and Cdk6, and Ki67 a marker of cell proliferation within the baboon fetal adrenal cortex during advancing stages of gestation. Fetal adrenal glands were obtained on days 60 (early), 100 (mid), and 160-170 (late) of gestation (term = 184 days). Mean (+/- s.e.) cyclin D1 mRNA levels, determined by RT-PCR and expressed relative to 18S rRNA, were similar at early (0.85 +/- 0.09) and mid (1.04 +/- 0.08) gestation, then decreased (P < 0.001, ANOVA) approximately 50% by late gestation (0.57 +/- 0.04). Cyclin E mRNA levels were also similar at early (2.03 +/- 0.07) and mid (1.63 +/- 0.31) gestation, and decreased by 70% (P < 0.001) in late gestation (0.53 +/- 0.09). Coinciding with the decrease in cyclin D1 and cyclin E, the percentage of Ki67 positive cells in the definitive zone decreased twofold (P < 0.01) between mid (28.2 +/- 3.6) and late (13.8 +/- 1.7) gestation. The cyclin D1 and cyclin E proteins, determined by immunocytochemistry, were expressed at high levels in the definitive zone of baboon fetal adrenal gland, where they decreased between mid- and late gestation. In contrast, immunocytochemical expression of the functionally important steroidogenic enzyme Delta(5)-3beta-hydroxysteroid dehydrogenase (3beta-HSD) became abundant in the definitive and transitional zones with advancing pregnancy. However, fetal adrenal Cdk2, Cdk4, and Cdk6 mRNA levels and protein immunoexpression were similar in the baboon fetal adrenal at early-, mid-, and late gestation. In summary, expression of cyclin D1, cyclin E, and Ki67 decreased, while 3beta-HSD expression increased, in the fetal adrenal cortex, particularly in the definitive zone, between mid- and late-baboon gestation. We propose that a developmental decline in cellular proliferation permits functional differentiation of fetal adrenal cortical cells, leading to increased production of steroid hormones important for placental estrogen synthesis and maturation of organ systems within the developing fetus.

This previous study was performed to compare the antero-posterior and medium-lateral displacements and the speed of displacement of the COP of healthy children and compare them to children with cerebral palsy type spastic diparesy. This study had the participation of 10 healthy children (6 girls and 4 boys), between 5 and 10 (7,6 +/- 2,1) years old, and other 10 children, all diagnosed as CP type spastic diparesy, (5 girls and 5 boys), between 5 e 10 (7,3 +/- 1,8) years old. To collect the data, we used the antero-posterior (AP) e medium-lateral (ML) displacements of the COP The parameters were compared by the variance analysis (ANOVA), and the post hoc test was Tukey's HSD. The level of significance adopted was p < 0,05. After the collect of data and statistical analysis, it was observed that the medium amplitude of the AP and ML displacements was higher in CP children, related to the healthy children: AP-F(I,18)= 1,66 (p = 0,002), ML-F1,18)= 3,36 (p = 0,002). As the amplitude, the speed of the displacements was also different among the children. The speed of the displacements, AP and ML was higher in CP children - F(1,36) = 0,009 (p = 0,00). There were no differences between the speed of AP and ML displacements, neither in the healthy children, nor in CP ones - F(1,36)= 0,00 (p = 0,87). The sensorial deficit found in CP children is an important issue in the increase of the amplitude and speed of displacement of the COP.

The purpose of this study was to compare different solutions for the storage of teeth and their influence on the enamel surface. The bond strength of bonded brackets on the enamel surface was determined by shear testing with an Instron measuring machine (feed rate 0.5 mm/min). Previously, these teeth had been stored in different solutions and their bond strengths were compared to the values of freshly extracted teeth. The statistical depiction was performed by means of descriptive and explorative data analysis. The analysis was carried out with the SPSS program. For group comparison purposes, the single-factor variance analysis (ANOVA) and the Post Hoc Test (Tukey HSD) were used. The level of significance was set at P < .05. The study showed that bond strength using formaldehyde fixation came closest to bond strength from freshly extracted teeth, so a formaldehyde solution should be used for storage. Only bond strength with teeth stored in thymol was significantly different (P = .02) from that observed with freshly extracted teeth.

The purpose of this study was to compare vascularization of the nasal mucous membrane among non-allergic, non-treated allergic and allergic patients treated with mometasone furoate, by means of the stereology method in quantitative analysis. Three groups of patients (GP), each containing 10 patients were examined. The first group (GP 1) had a negative inhalatory allergen test while the second (GP 2) and third (GP 3) group both had positive results with the same test. GP 3 included allergic patients treated with mometasone furoate for 15 days before analysis, when a small piece of the nasal mucous membrane was taken from the frontal pole of the lower nasal shell. The specimens were examined immunohistochemically for expression of CD31 and VEGF-C. Vascular phase was determined by using length density (L(v)). Differences in CD31 and VEGF-C expression were compared using one-way ANOVA and Tukey HSD post-hoc tests. CD31 expression in GP 1 had significantly lower values than in the GP 2 and GP 3 groups (p < 0.001). VEGF-C expression in GP 1 was significantly lower than in GP 2 (p = 0.007), but not in GP 3 (p = 0.292). We have shown that 15-day treatment with mometasone furoate results in a significant reduction of the density of vascular elements in allergic patients.

Riluzole is the only FDA approved drug for the treatment of amyotrophic lateral sclerosis. Riluzole is assumed to be mainly metabolized by the liver cytochrome CYP1A2 and by the extra-hepatic cytochrome CYP1A1. CYP1A2 and CYP1A1 genetic polymorphisms are known, but their relationship to riluzole metabolism in ALS patients has not been investigated. The aim of this study was to determine whether the polymorphisms of the CYP1A2 and the CYP1A1 genes in ALS patients are associated with riluzole metabolic profiles. Thirty-two patients with a diagnosis of probable or definite ALS and who were on riluzole, participated in the study. Trough and peak plasma riluzole levels were measured using analytical chromatography-mass spectrometry methods. Association of the genotypes of the SNPs spanning the CYP1A1 and CYP1A2 genes (including one SNP in the intergenic region) with mean riluzole peak and trough levels was studied using ANOVA and Tukey's HSD. The mean peak riluzole level was 202+/-111 ng/ml and mean trough level 54.3+/-37.5 ng/ml. Our data do not support any association of the four CYP1A1 and CYP1A2 polymorphisms with the riluzole metabolic profile. In conclusion, genetic variations in CYP1A1 and CYP1A2 genes do not seem to influence riluzole levels. Further work is needed to better understand the genetic regulation of CYP1A enzymes and their role in riluzole metabolism.

The aim of this study was to evaluate the effects of irradiance, light emission wavelength, and heating of different light curing units on the Knoop and Vickers hardness of a hybrid composite resin. The specimens were irradiated during 40 s with ten different light curing units, LEDs, and halogen lights. The spectral emission of each light curing unit was assessed by a spectrometer, the irradiance was measured by two commercial radiometers, and the heating measured with a thermocouple. After 48 h of storage in a dark recipient under a 100% humidity condition, the Knoop and Vickers hardness tests were carried out. The hardness results were analyzed by ANOVA, and Tukey HSD test (p < 0.05). The results showed that the surface hardness of the composite resin depends not only on the irradiance, but strongly on the emission wavelength and heating of the light curing units. It was observed, a linear correlation between the conversion degree and radiant exposure. In addition, it is suggested that the well known base to top surface hardness ratio convention of 80-90% is not appropriate to evaluate curing efficiency of composites, since the top surface is not always sufficiently polymerized.

The purpose of this study was to evaluate the influence of irradiation time on volumetric change and flexural properties of flowable resins. Four commercially available flowable resins were employed. For volumetric shrinkage measurement, resin pastes were inserted into a mold (2 mm in height, 4 mm in diameter) and put into a water-filled dilatometer. This was followed by light irradiation for 10, 20, or 30 seconds at 600 mW/cm2. Volumetric shrinkage of the specimens was then determined from the change in the height of water meniscus, and the percentage volumetric change thereof was calculated. For flexural strength measurement, resin pastes were filled into a stainless steel mold (25 x 2 x 2 mm), and the middle one-third of the specimen was first irradiated. The remaining two-thirds were irradiated under the same irradiation conditions as volumetric shrinkage measurement. After 24-hour storage in 37 degrees C water, three-point flexural tests were performed with a span length of 20 mm at a crosshead speed of 1.0 mm/min. One-way ANOVA followed by Tukey's HSD test were used for statistical analysis. For all materials tested except Estelite Flow Quick, both volumetric shrinkage and flexural strength increased with longer light irradiation time. Results of this study indicated that both volumetric shrinkage and flexural properties were influenced by light irradiation time and resin composite type.

Extracorporeal circulation (ECC) is used in cardiac surgery for cardiopulmonary bypass as well as in ventricular assist devices and for extracorporeal membrane oxygenation. Blood contact with the artificial surface and shear stress of ECC activates platelets and leukocytes resulting in a coagulopathy and proinflammatory events. Blockers of the platelet glycoprotein (GP) IIb/IIIa (CD41/CD61) can protect platelet function during ECC, a phenomenon called "platelet anaesthesia", but may be involved in post-ECC bleeding. We hypothesized that the new selective phosphoinositide 3-kinase p110beta inhibitor TGX-221 that inhibits shear-induced platelet activation without prolonging the bleeding time in vivo may also protect platelet function during ECC. Heparinized blood of healthy volunteers (n = 6) was treated in vitro with either the GP IIb/IIIa blocker tirofiban, TGX-221 or as control and circulated in an ECC model. Before and after 30 minutes circulation CD41 expression on the ECC-tubing as measure for platelet-ECC binding and generation of the platelet activation marker beta-thromboglobulin were determined using ELISA. Platelet aggregation and platelet-granulocyte binding were analysed in flow cytometry. After log-transforming the data statistical evaluation was performed using multifactor ANOVA in combination with Tukey's HSD test (global alpha = 5%). Tirofiban and TGX-221 inhibited platelet-ECC interaction, platelet aggregation and platelet-granulocyte binding. Tirofiban also inhibited ECC-induced beta-thromboglobulin release. The observed inhibition of platelet-ECC interaction and platelet activation by tirofiban contributes to explain the mechanism of "platelet anaesthesia". TGX-221 represents a promising alternative to GP IIb/IIIa blockade and should be further investigated for use during ECC in vivo.

This study investigated the leakage pathway of facial and lingual Class V cavities restored with different flowable resin composites bonded with one bonding agent by examining the resin/dentin interface. Forty Class V cavities were etched with 37% phosphoric acid gel; Single Bond dental adhesive was applied, then the cavities were randomly divided into four groups (n=10). Three groups were restored with one of three flowable resin composites (Grandio Flow, Filtek Flow and Admira Flow). The fourth group was restored with Z250 (hybrid resin composite) to serve as a control. The specimens were then placed in 50% w/v silver nitrate solution for 24 hours and immersed in a photodeveloping solution for eight hours. Thereafter, the specimens were sectioned bucco-lingually, polished, mounted on stubs, gold sputter coated and examined by scanning electron microscope. Silver particle penetration length with and without gap formation was measured directly on the scanning electron microscope monitor and calculated as a percentage of the total length of the cut dentin surface that was penetrated by silver nitrate. The data were analyzed with one-way ANOVA and Tukey HSD test. The groups restored with Filtek Flow and Admira Flow showed a microleakage pattern where silver nitrate penetration was observed with gap formation at the tooth/restoration interface and Filtek Flow recorded significantly higher leakage than Admira Flow. Grandio Flow showed similar marginal adaptation to Z250 resin composite with no gap formation at the interface. However, silver ions had penetrated beneath the resin-impregnated layer in cavities restored with Grandio Flow and Z250, indicating nanoleakage occurred. This study suggests that volumetric shrinkage in resin composites remains a problem. Although some new technologies are trying to solve the problem of composite shrinkage, the bonding system used in this study did not achieve perfect sealing at the restoration/dentin interface. This might affect durability of the bond to dentin.

This study evaluated the tensile bond strengths of three adhesive/composite core materials to bovine dentin using three different curing units. Bovine dentin surfaces were ground with 600-grit SiC paper. Bonding area was demarcated with a vinyl tape (4-mm-diameter hole). Three adhesive/composite core systems--S6054 (experimental), UniFil Core, and Clearfil DC Core Automix--were used with three curing units--Curing Light XL3000 (quartz-tungsten-halogen), Hyper Lightel (high-power quartz-tungsten-halogen), and LEDemetronl (blue light-emitting diode)--according to manufacturers' instructions. After 24 hours of storage in water at 37 degrees C, tensile bond strengths were measured at a crosshead speed of 2 mm/min. Results were statistically analyzed with one-way ANOVA and Tukey's HSD test (p < 0.05). Highest tensile bond strength was obtained using Clearfil DC Core Automix with Hyper Lightel.
