In 30 cases of disease of the knee joint (9 cases of rheumatoid arthritis, 14 arthroses and 7 cases of posttraumatic postoperative synovitis), the quantitative changes of the four lysosomal enzymes (acid phosphatase, lactic acid dehydrogenase, cathepsin and proteins) was observed in the course of intra-articular treatment with Gordox. It was evident from the analysis of the enzyme determinations conducted within 5--30 days at various times, and from the clinical results of the treatment, that in the cured or improved cases, all four enzymes were reduced (6 patients), whereas, in 16 cases, three of the enzyme levels had decreased. The enzyme titer remained unchanged in those patients who had been treated without success. Although these results do not as yet allow any final conclusions with regard to the mechanisms of action of Gordox administered intraarticularly, they do indicate that further and even more thorough studies in this direction should be conducted.

The activity of gold sodium thiomalate (GST) given i.m. to adjuvant-induced polyarthritic rats was studied alone or in combination with active doses of aspirin, indomethacin and hydrocortisone. In addition to paw volume and body weight changes, erythrocyte sedimentation rate, serum albumin/globulin and gold levels as well as plasma activities of beta-glucuronidase, acid phosphatase, lysozyme and lactic acid dehydrogenase were measured. In prophylactic studies the beneficial activity of GST was unaffected by aspirin, suggesting a positive drug interaction, but additive with indomethacin or hydrocortisone for the 1st but not 2nd lesion of the disease. These results were closely correlated with increased serum gold levels. Similar clinical findings were observed in therapeutic studies except that a positive drug interaction occurred between GST and hydrocortisone. Unlike in the prophylactic experiments, serum gold levels were unaffected by any of the agents tested in the therapeutic studies.

Histological and histochemical studies were carried out on the gastro-intestinal mucosa of three experimental cows (roughages/maize silage) and three control animals. The animals were slaughtered on termination of the long-term trial. Mucosa samples were taken, for further study, from the rumen, the duodenum, the jejunum, the large intestine and the appendix. Histochemical analysis did not reveal any essential differences in the activities of non-specific esterase, alkaline and acid phosphatase and lactic acid dehydrogenase in the mucosa of the rumen, the large and the small intestine and the appendix of both the experimental animals and the controls. The experimental animals were found to exhibit a higher rate of glutamate-dehydrogenase activity in the ruminal mucosa and in the mucosa of the large and small intestine. A higher succinate dehydrogenase activity was observed in the ruminal mucosa of the experimental animals, relative to that of the controls, while the activity in the intestinal mucosa was decreased. Only slight changes were noted in the activity of the enzymatic systems tested. Electron microscopic studies did not reveal any differences in the ultrastructure of the epithelial cells of the ruminal mucosa in both the experimental animal and the controls.

The Glycolipoprotein (GLP) of various virulent leptospires was extracted, purified and identified. The GLP was combined with fluorescence rhodamine B and the colloidal gold as the probe. Meanwhile, the bovine aortic endothelial cell (EN) was isolated and cultured in the medium 199 "Nissui". The GLP-rhodamine B and GLP-colloidal gold probe were added to cultured EN and incubated at 4 degrees C for 2 h then at 37 degrees C for 2 h. The interaction between GLP-rhodamine B, GLP-CG probe and EN were observed and photography by fluorescence microscopy (Leitz) and scanning electronmicroscopy. The releasing rate of lactic acid dehydrogenase (LDH), acid phosphatase (AcP) and protein from the EN were detected. The results indicated that the GLP of Leptospira interrogans serovar Lai, strain 017 attached to EN and a endocytosis was happened subsequently. The LDH and AcP releasing rate were significantly higher than control and the cell protein was much less than control. The results showed that the GLP of L. interrogans strain 017 had the cytotoxin effect through a specific attachment, endocytosis in EN. The endocytosis probably is a receptor-mediated endocytosis.

The screening of autopsy specimens, vaginal, buccal, and rectal swabs, for the presence of seminal fluid in rape homicide investigations utilizing classical techniques can lead to erroneous results. In the absence of spermatozoa, techniques are needed which can help to identify seminal fluid. This report illustrates the use of a multi-enzyme electrophoretic approach identifying seminal acid phosphatase (SAP) and lactic acid dehydrogenase (LDH-X) as an initial screening procedure. Subsequent analyses for the presence of acid and alkaline phosphatase (semiquantitative) yield information which can help identify false-positive SAP's. Additionally, salivary amylase can be tentatively identified using the same multi-enzyme procedure which informs the investigator of possible salivary contamination of the sample and possible erroneous PGM results. Statistics utilizing the multi-enzyme approach in case work are also presented.

Inhalation of manganese oxide (Mn dose, 879.0 micrograms/m3) for 2 h reduced the total number of alveolar macrophages obtained by endotracheal lavage in pulmonary cell populations, slightly reduced cellular viability, and reduced both phagocytic capability and total protein in sonicated pulmonary cells. Increases in intracellular adenosine triphosphate and acid phosphatase specific activity were also exhibited by the pulmonary cells, but sonicated cells obtained from the exposed mice showed no change in lactic acid dehydrogenase specific activity. A slight increase in extracellular protein in the fluid phase of the lavage suspension was observed after manganese oxide exposure.
