Perfusion of the rat liver in situ for 150 min provides for maintaining optimal values of acid-base balance for the following indexes: surplus of bases, content of standard bicarbonate, buffer bases pH, pO2, pCO2, HbO2, the level of bile secretion, content of lactate, pyruvate, ATP, ADP, that evidences for a high functional activity in the tissue. Introduction of dinitro-ortho-cresol (DNC) into the perfusion liquid causes development of acidosis. DNC results in dissociation of oxidative phosphorylation: the content of ATP and intensity of inorganic phosphorus utilization decrease, oxygen uptake intensifies. A compensatory increase in the glycolysis intensity directed to maintaining the level of macroergs under these conditions is is pronounced in the intensified uptake of glucose, in a rise in the content of lactate in perfusate and an increase in the pyruvate kinase activity in the liver. The redox state of NAD-pairs (ratio of [NAD+] : [NADN] calculated from the content of redox metabolites and the equilibrium constant for the lactate dehydrogenase system shifts toward an increase in the reducing properties of hepatocytes cytoplasm. The phosphate potential value calculated from the ratio [ATP] : [ADP] - [Pinor] lowers under conditions of the experiment.

Activity of enzymes catalizing bioenergetic processes of substance transport through cell membranes, adenosine triphosphatase and para-nitrophenyl phosphates, activity of certain enzymes of carbohydrate metabolism, lactate dehydrogenase and glucose-6-phosphate dehydrogenase, as well as to 5'-nucleotidase taking part in nucleic metabolism were determined in the pancreas of thyreoidectomized rats. Simultaneously the content of lactic acid, phosphorus, potassium and sodium, which immediately related to activation of the mentioned enzymes, was determined in the pancreas. In thyroidectomized rats the activity of Mg2+, Na+, K+-ATPase, Na+, K+-ATPase and lactate dehydrogenase in the pancreas increases, that of glucose-6-phosphate dehydrogenase, para-nitrophenylphosphatase and 5-nucleotidase decreases, the content of lactic acid, potassium, sodium and phosphorus increases.

Normal values for 13 chemical constituents of plasma were estimated from results for 837 presumably healthy children. Ninety microliters of specimen was analyzed for lactate dehydrogenase, aspartate aminotransferase, alkaline phosphatase, inorganic phosphorus, total calcium, total cholesterol, total proteins, albumin, uric acid, urea nitrogen, alanine aminotransferase, total bilirubin, and glucose. We used two Abbott ABA-100 Bichromatic Analyzers interfaced directly to the ABA Data Management System. For each test age- and sex-related variations were assessed and normal values were estimated for six different age groups.

In order to verify the influence of sampling time on blood constituents, populations of supposedly healthy subjects were grouped according to age, sex, deviation from their ideal weight, state of fasting or nonfasting, and time of sampling. Each fasting subject in one group underwent two samplings during the course of a morning: the first at 08.00 and the second between 09.00 and 12.00. In the second group, the first was taken at 13.00, and the second between 14.00 and 16.00. Subjects in the second group had eaten a standard meal of 700 calories at 12.00. Differences between the paired samples from a given individual are discussed with respect to the time of sampling for plasma urea, creatinine, proteins, albumin, calcium, sodium, potassium, cholesterol, uric acid, chloride ions, phosphate, bilirubin, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, creatine phosphokinase, alkaline phosphatase, hemoglobin and erythrocyte and leukocyte counts. Variations due to the time of sampling were large for phosphorus, bilirubin, and leukocyte count.

In serial studies of hepatic function in rats after 70% partial hepatectomy, quantitative changes were found in several of the serum components used clinically to assess liver status. The activities of the following enzymes were found to increase: gamma-glutamyl transpeptidase and lactic dehydrogenase were maximal 6 h postoperatively, while glutamic oxaloacetic transaminase and alkaline phosphatase reached peak values at 24 and 48 h respectively. Albumin levels were found to be relatively constant during the study; however, total protein concentration was lowest 6--12 h postoperatively, paralleling a decrease in globulin concentration. Bilirubin levels were elevated to 4x normal within 12 h after surgery. After partial hepatectomy calcium and phosphorus concentrations were significantly decreased at 24 and 12 h respectively. With the exception of alkaline phosphatase, the activities of all serum components measured returned to normal levels by 1 week after surgery; the alkaline phosphatase concentration continued to be elevated 2 weeks postoperatively.

A study of cardiovascular risk factors in middle-aged twin men provided an opportunity to test for genetic variability in the SMA 12/60 (Technicon) battery of clinical chemistry tests. Classical twin methodology was used to analyze the variation of monozygotic and dizygotic twins. In addition, frequency of co-twin contact was used to control for effects of differences in shared environment. Genetic variability played a definite role in controlling four of the 11 reported tests: one-hour serum glucose, serum urea nitrogen, uric acid, and bilirubin. No genetic variation was found for lactate dehydrogenase, phosphorus, and alkaline phosphatase. Significantly higher means for calcium, total protein, albumin, and aspartate aminotransferase in monozygotic twins precluded any statement about heredity and environment for these tests.

The precision and accuracy of 21 methods in clinical chemistry (sodium, potassium, calcium, chloride, phosphorus, iron, copper, bilirubin, uric acid, urea, creatinine, total serum protein, triglycerides, glutamate-oxalacetate-transaminase, glutamate-pyruvate-transaminase, alpha-hydroxybutyrate-dehydrogenase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyl-transpeptidase, acid phosphatase, amylase) were studied after diluting samples 1 : 2 and 1 : 4. Dilution of 1 : 2 did not show any significant effect on the precision and accuracy of the studied methods. In case of chloride, phosphorus, iron, bilirubin, uric acid, urea, creatinine, total serum protein, triglycerides, glutamate-oxalacetate-transaminase and alkaline phosphatase reliable results were obtained also after 1 : 4 dilution.

A sample of n = 836 male students between 9 and 16 years from Barcelona was analysed for quantitative variations of various hematological characters during growth. The results were compared with a sample of n = 194 adult males between 20 and 45 years. No statistically significant variations were found concerning the levels of calcium, ureic nitrogen, glutamate-oxal-acetat-transaminase (s-GOT), serum albumin and total serum protein. Against that, marked and statistically significant age variations were seen for the levels of alkaline serum phosphatase, glucose, anorganic phosphorus, cholesterol, and uric acid. These variations are without doubt associated with the metabolic processes during growth. Unexplained remain the age variations concerning the bilirubin and lactate dehydrogenase levels. Out of the serum protein fractions only the alpha1- and gamma globulins showed some age variations: The alpha1-fraction increases between 10 and 16 years, the gamma fraction decreases between 9 and 14.

Forty-seven male Macaca mulatta, 3 to 4 kg weight, were inoculated intravenously or subcutaneously with various doses of yolk sac-grown Rickettsia rickettsii. Thirty-four macaques became febrile and exhibited signs of infection ranging from transient illness with a few days of fever to severe illness with subsequent death. The rash appeared more frequently in the macaques inoculated subcutaneously. Febrile macaques that survived had leukocytosis, with concomitant neutrophilia. Febrile macaques that died had, in addition, marked terminal leukopenia and thrombocytopenia. Packed cell volume of all febrile macaques decreased. In almost all of the febrile macaques, there were increased serum urea nitrogen, glutamic-oxaloacetic transaminase, and lactate dehydrogenase and decreased total serum protein and amylase concentrations. A few febrile macaques had increased bilirubin values and decreased sodium, chloride, phosphorus, and alkaline phosphatase concentrations. Changes did not occur in serum glucose, potassium, calcium, and glutamic-pyruvic transaminase values. The experimental form of Rocky Mountain spotted fever in the macaque provides a subhuman primate model for studying the pathophysiology of this disease.

The values of a number of biochemical variables have been studied before and after a 50-gram load of glucose orally. Reductions which were statistically significant were found for sodium, potassium, urea, total protein, albumin, calcium, phosphorus, urate, bilirubin, alkaline phosphatase, but not for bicarbonate, creatinine, creatine kinase, lactate dehydrogenase, aspartate aminotransferase, cholesterol, triglyceride or chloride. The magnitude of the changes was generally not great, but could be clinically appreciable. The differences may need to be taken into account in comparing population studies.
