Histochemical techniques have been employed to characterize enzymatic activity in the mesocoxal muscles of the cockroach, Periplaneta americana. Through our studies of the enzymes myosin-ATPase,
NADH
reductase, succinic dehydrogenase (SDH), and
lactic dehydrogenase
(
LDH
), we were able to classify fibers within these muscles according to criteria established for muscle fibers of vertebrates. Many of the mesocoxal muscles possess two different and distinct populations of fibers, whereas the remaining muscles are homogeneous with respect to their constituent fibers. The data presented here indicate biochemical heterogeneity for muscles of differing structural and functional features and possible neurotrophic influences upon oxidative enzymes and myosin-ATPase isozymes.

The case of a 35 years-old man, with chronic proximal muscle atrophy in which at the muscle biopsy tubular aggregates were found by histochemistry procedures is reported. The tubular aggregates stained positive with the modified Gomori trichrome, haematoxylin-eosin,

-diaphorase, non specific esterases, phosphorylase, P.A.S., oil red O and
lactate dehydrogenase
. They did not show in the routine and acid pre-incubated ATPase, acid and alkaline phosphatases and succinate dehydrogenase. Only found in type II fibers. A brief discussion about the pathogenesis and function of the tubular aggregates is made. The authors believe that the tubular aggregates in this case are secondary to prolonged use of phenobarbital and diphenylhydantoin, associated with the basic denervation process and alcohol abuse.

Examination of selected oxidoreductases (succinate dehydrogenase, mitochondrial glycerol-3-phosphate dehydrogenase,

tetrazolium reductase) in the rat gastric mucosa revealed diurnal fluctation of enzyme activities with the most marked manifestation in succinate dehydrogenase. The maximum enzymatic activity found at 18.00 h and 24.00 h) points to the highest oxidoreductase capacity in the parietal cells just at the time when a rat usually expresses spontaneously the highest interest in food intake. The high activity of succinate dehydrogenase and other enzymes at that time is very likely the expression of a "fixed" metabolic adaptation of the parietal cells to the elevated production of hydrochloric acid, in connection with its role in the digestion of food in the stomach. The low enzymatic activity of most rat parietal cells during the day may represent the picture of "a resting afunctional".

Muscle samples were obtained from the gastrocnemius of 17 female and 23 male track athletes, 10 untrained women, and 11 untrained men. Portions of the specimen were analyzed for total phosphorylase,
lactic dehydrogenase
(
LDH
), and succinate dehydrogenase (SDH) activities. Sections of the muscle were stained for myosin adenosine triphosphatase,
NADH2
tetrazolium reductase, and alpha-glycerophosphate dehydrogenase. Maximal oxygen uptake (VO2max) was measured on a treadmill for 23 of the volunteers (6 female athletes, 11 male athletes, 10 untrained women, and 6 untrained men). These measurements confirm earlier reports which suggest that the athlete's preference for strength, speed, and/or endurance events is in part a matter of genetic endowment. Aside from differences in fiber composition and enzymes among middle-distance runners, the only distinction between the sexes was the larger fiber areas of the male athletes. SDH activity was found to correlate 0.79 with VO2max, while muscle
LDH
appeared to be a function of muscle fiber composition. While sprint- and endurance-trained athletes are characterized by distinct fiber compositions and enzyme activities, participants in strength events (e.g., shot-put) have relatively low muscle enzyme activities and a variety of fiber compositions.

was studied in the rat perineurium 10, 20, 30, and 90 days post partum. The effect of under-nutrition was shown by a lack of enzyme activity in rats 30 days post partum in contrast to control animals, 30 days also corresponds to the time when, in normal rats, the perineurial diffusion barrier to exogenous macromolecules becomes established. In undernourished rats this diffusion barrier does not appear. At 90 days post partum raised enzyme activities indicate a high metabolic turnover in the perineurium even in animals with undernutrition.

Male guinea pigs were exposed to nitrogen dioxide (2 mg/m3) during 180 days (8 hours a day). Long-term exposure induced thickening of the corneal layer of the epidermis as well as inflammatory infiltrations in the proper skin. Formation of the binary complex between the reduced coenzyme nicotinamide adenine dinucleotide (
NADH
) and pig skeletal muscle
lactate dehydrogenase
(LDH,

) has been investigated by calorimetric and equilibrium dialysis techniques in 0.2 M potassium phosphate buffer (pH 7.0) at various temperatures. Analysis of thermal titration curves at two temperatures (25 and 31.5 degrees) shows that the experimental enthalpy data can be rationalized assuming four independent and equivalent binding sites for the tetrameric enzyme. Binary complex formation is characterized by a negative temperature coefficient, delta cp, of the binding enthalpy, which amounts to -1300 plus or minus 53 cal/(deg mol of LDH) in the temperature range of 5-31.5 degrees. Despite the slightly smaller standard deviation resulting when polynomial regression analysis of the second degree is applied to the temperature dependence of the enthalpy values, binding enthalpies seem to be adequately represented in the temperature range studied by the equation delta H = -1.3T + 2.3, kcal/mol of LDH, T referring to the temperature in degrees C. By combination of the results obtained from equilibrium dialysis and calorimetric studies a set of apparent thermodynamic parameters for binding of
NADH
to LDH in 0.2 M potassium phosphate buffer at pH 7 has been established.

To determine the molar absorptivities of
beta-NADH
and beta-NAD at 260 nm, we purified the reduced form of the coenzyme by repeated anion-exchange chromatography. A
NADH
preparation was so obtained for which the 260 nm/340 nm absorbance ratio was 2.265. When calculated with epsilon 340
beta-NADH
= 6.22 times 10-3 for
beta-NADH
at 260 nm and 25 degrees C, a molar absorptivity of 14.1 times 10-3 liter - mol minus 1 - cm minus 1 resulted from this quotient. By use of the
lactate dehydrogenase
or glycerol-3-phosphate dehydrogenase assay, respectively, and referring to the new absorption coefficient for
NADH
at 260 nm, the molar absorptivity of beta-NAD at 260 nm and 25 degrees C was established to be 17.4 times 10-3 liter - mol minus 1 - cm minus 1. The values observed are lower than those reported thus far.
